摘要
目的:基于网络药理学方法,初步探究潜降汤在高血压治疗方面的作用机制。方法:通过检索中药系统药理学数据库和分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)以及中国知网,得到潜降汤全方共10味中药的活性化合物,在此基础上构建"化合物-靶点"网络。依托治疗靶点数据库(therapeutic target database,TTD),搜集高血压疾病靶点,结合化合物预测靶点,进而基于蛋白质-蛋白质相互作用(protein-protein interaction,PPI)分析、基因本体(gene ontology,GO)功能富集分析和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析,探究潜降汤治疗高血压的作用机制。结果:共得到潜降汤化合物69个,预测靶点580个,高血压相关疾病靶点658个,化合物-疾病共有靶点90个。进行PPI分析,得到90个体现药物与疾病直接或间接作用的靶点蛋白;富集分析中得到显著富集的22个GO条目,其功能主要有大分子的相互作用、细胞结构组成等;得到32条KEGG通路,包括恶性肿瘤通路、肾细胞癌通路和催乳信号途径等。结论:本研究通过网络药理学和生物信息整合分析方法,探索了潜降汤治疗高血压的作用机制,初步推测其中发挥治疗作用的主要靶点包括AKT1、SRC和CXCL12等,且与质膜组成、Janus激酶/信号转导与转录激活子(Janus kinase-signal transducer and activator of transcription,JAK-STAT)级联和趋化因子信号通路等有关,但具体作用机制尚需实验验证。
OBJECTIVE:To preliminarily explore the mechanism of Qianjiang decoction in the treatment of hypertension based on network pharmacology.METHODS:Traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)and CNKI were retrieved to collect 10 kinds of active compounds of traditional Chinese medicine contained in Qianjiang decoction,on this basis,the Compound-Target network was constructed.By means of the therapeutic target database(TTD),the targets of hypertension were collected.Compound target network and compound putative target-hypertension target network were applied for protein-protein interaction(PPI),gene ontology(GO)function enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.The mechanism of Qianjiang decoction in the treatment of hypertension was explored.RESULTS:A total of 69 Qianjiang decoction compounds,580 predicted targets,658 hypertension-related disease targets and 90 compound-disease targets were obtained.Through PPI analysis,90 target proteins with direct or indirect effects of drugs and diseases were obtained.Totally 22 GO items were significantly enriched in the enrichment analysis,the functions mainly included macromolecule interaction and cell structure composition.Totally 32 KEGG pathways consisted in cancer pathway,renal cell cancer pathway and breast stimulating signal pathway.CONCLUSIONS:This study explored the mechanism of Qianjiang decoction in the treatment of hypertension through the method of network pharmacology and biological information integration analysis.It is preliminarily speculated that the main therapeutic targets are AKT1,SRC and CXCL12,which are related to plasma membrane composition,Janus kinase-signal transduction and activator of transcription(JAK-STAT)cascade and chemokine signaling pathway,yet the specific mechanism needs to be verified by some experiments.
