摘要
目的:探究活血醒脑方对颅脑损伤大鼠模型神经细胞凋亡的影响。方法:选40只6周龄SPF级大鼠体质量200±30g,采用改良的Feeney’s自由落体打击装置造重度颅脑损伤大鼠模型,并随机分为4组,空白对照组(control)、模型组(model)、低剂量活血醒脑方组(Low HXXNF)和高剂量活血醒脑方组(High HXXNF);使用TUNEL法原位检测损伤区神经细胞的凋亡情况;使用Western blot检测大鼠颅脑组织Bcl-2、Bax、Caspase-3、Caspase-9蛋白表达量;使用流式细胞仪检测细胞凋亡率。结果:相比空白对照组,模型组大鼠神经细胞的凋亡数目、凋亡细胞指数(ACI)、Caspase-3、Caspase-9和Bcl-2蛋白表达量显著增加,Bax蛋白表达量显著降低,且差异有统计学意义(P<0.05);相比模型组,活血醒脑方组大鼠神经细胞的凋亡数目、凋亡细胞指数、Caspase-3、Caspase-9和Bcl-2蛋白表达量显著降低,且高剂量活血醒脑方组上述指标显著低于低剂量活血醒脑方组,差异有统计学意义(P<0.05),Bax蛋白表达量显著升高,且高剂量活血醒脑方组Bax蛋白表达量显著高于低剂量活血醒脑方组,差异有统计学意义(P<0.05)。结论:活血醒脑方可以通过调节细胞凋亡因子抑制颅脑损伤大鼠模型神经细胞凋亡,且在一定浓度范围内呈浓度依赖性。
Objective:To explore the effect of Huoxue Xingnao Recipe on nerve cell apoptosis in rat models with traumatic brain injury.Methods:A total of 406-week-old SPF rats weighing 200±30 g were selected,and rat models of traumatic brain injury were made by modified Feeney’s free fall device and were randomly divided into four groups,including controlled group,model group,low-dose Huoxue Xinnao Recipe group and high-dose Huoxue Xingnao Recipe group.TUNEL in situ test method was used to detect apoptosis of injured nerve cells.Western blot was used to detect the protein expression levels of Bcl-2,Bax,Caspase-3 and Caspase-9 in rat brain tissue,and flow cytometry was used to detect the apoptosis rate of cells.Results:Compared with the blank controlled group,the number of apoptosis,apoptotic cell index(ACI)and protein expression levels of Caspase-3,Caspase-9 and Bcl-2 in the model group were significantly increased,and the protein expression level of Bax was significantly decreased(P<0.05).Compared with the model group,the number of apoptosis,apoptotic cell index and protein expression levels of Caspase-3,Caspase-9 and Bcl-2 in the Huoxue Xingnao recipe groups were significantly decreased,and the above-mentioned indicators in the high-dose Huoxue Xingnao recipe group were significantly lower than those in the low-dose Huoxue Xingnao recipe group(P<0.05),and the Bax protein expression level was significantly increased,and the Bax protein expression level in the high-dose Huoxue Xingnao Recipe group was significantly higher than that in the low-dose Huoxue Xingnao Recipe group(P<0.05).Conclusions:Huoxue Xingnao Recipe can inhibit the apoptosis of rats models of traumatic brain injury by regulating apoptosis factors,and in a certain concentration range,it is concentration-dependent.
