摘要
为研究干酪乳杆菌对发育期腹泻模型大鼠回肠黏膜结构及黏蛋白2(MUC2)含量的影响,利用苏木精-伊红(HE)染色法、过碘酸雪夫(PAS)染色法、蛋白免疫印迹(Western-blot)及RT-PCR方法,以36只4周龄SD雄性大鼠为试验对象,随机分成6组,每组6只。采用5%硫酸镁溶液灌胃构建腹泻模型,10~8 CFU/mL干酪乳杆菌(LcZhang)液灌胃进行预防治疗,生理盐水灌胃作为空白对照。结果表明:干酪乳杆菌预防(6 h)组回肠V/C值、杯状细胞数量、MUC2蛋白含量及MUC2 mRNA的表达量比腹泻(6 h)组分别增加了10.89%、9.28%、26.44%和66.67%(P=0.00,P<0.01,P<0.01,P<0.05);干酪乳杆菌预防(48 h)组回肠V/C值比腹泻(48 h)组显著增加了12.20%(P=0.00)。以上结果显示,饲用干酪乳杆菌进行预防能够显著提高腹泻大鼠回肠V/C值及杯状细胞数量,促进MUC2的合成,表明干酪乳杆菌可以有效维护4周龄大鼠肠道健康,减轻腹泻对大鼠肠道黏液屏障的损害,加强肠道免疫功能。
In order to study the effect of Lactobacillus casei on ileal mucosal structure and MUC2 content of rats with developmental diarrhea,36 male 4-weed-old Sprague-Dawley rats were randomly divided into 6 groups with 6 in each group.Hematoxylin-eosin staining,Periodic acid Schiff staining,Western blot and RT-PCR methods were adopted in this study.In the experiment,a diarrhea model was established by intragastric administration of 5%magnesium sulfate solution and 10~8 CFU/mL Lactobacillus casei Zhang(LcZhang)solution was intragastrically administered for prevention and treatment.Saline treatment only was administered as a control.The results showed that:The ileal V/C value,number of goblet cells,MUC2 content and expression of MUC2 mRNA in group of the Lactobacillus casei prevention group(6 h)were respectively increased by 10.89%,9.28%,26.44%,66.67%(P=0.00,P<0.01,P<0.01 and P<0.05),compared with the group of diarrhea group(6 h);The ileal V/C value of group in the Lactobacillus casei prevention group(48 h)was significantly increased by 12.20%(P=0.00),compared with the diarrhea group(48 h).The above results showed that the prevention of Lactobacillus casei significantly increased the ileal V/C value and the number of goblet cells in diarrhea rats,and promoted the synthesis of MUC2,indicating that Lactobacillus casei could effectively maintain the intestinal health of 4-week-old rats,reduce the damage of diarrhea on intestinal mucus barrier and strengthen intestinal immune function.
