摘要
忍冬(Lonicera japonica Thunb.)是忍冬科忍冬属半常绿性藤本植物,其未开放的花蕾或初开放的花经干燥处理后称为金银花,是药食两用品。在全球气候变暖加剧干旱风险,以及“中药材上山入林”政策的双重背景下,忍冬种质资源抗旱性评价对耐旱种质培育具有重要意义。本研究利用0%、5%、10%、15%、20%、25%六个梯度浓度的PEG-6000溶液模拟干旱条件进行抗旱处理,发现10%浓度PEG-6000是评价忍冬种质萌发期发芽指标的适宜浓度,15%浓度PEG-6000是评价忍冬种质苗期形态指标的适宜浓度。与0%浓度PEG-6000条件下相比,在10%浓度PEG-6000条件下,142份忍冬种质的发芽势下降幅度达75.34%、发芽率下降幅度为42.38%,均表现出显著的下降幅度;在15%浓度PEG-6000条件下,142份忍冬种质的根长下降幅度为8.24%、苗高下降幅度为34.38%,均表现出显著的下降幅度,表明这些种质间抗旱能力存在显著差异。利用相关性分析、主成分分析、隶属函数分析、聚类分析对忍冬萌发期和苗期抗旱性进行综合评价,筛选出高抗旱型种质6份、抗旱型种质9份、中抗型种质73份、敏感型种质41份、高敏感型种质13份。研究结果为干旱与非干旱地区忍冬精准引种栽培、抗旱种质选育及抗旱分子调控网络解析提供了坚实的材料基础。
Lonicera japonica Thunb.,a semi-evergreen liana,produces the dried medicinal material Lonicerae Japonicae Flos,derived from either unopened flower buds or newly opened flowers.Recognized as a dual-purpose medicinal and edible substance in pharmacopeial standards,this species holds significant agricultural and pharmaceutical value.Against the backdrop of increasing drought risks due to global climate change and the implementation of China′s“Chinese Medicinal Materials Mountain-Forest Cultivation”policy,assessing the drought resistance of L.japonica germplasm resources holds significant implications for breeding drought-tolerant varieties.In this study,drought stress was simulated using polyethylene glycol-6000(PEG-6000)solutions at six concentration gradients(0%,5%,10%,15%,20%,25%)to simulate water-deficit conditions.The results determined that 10%PEG-6000 was the optimal concentration for evaluating germination-stage traits,whereas 15%PEG-6000 was most suitable for assessing seedling-stage morphological characteristics.Compared to the control(0%PEG-6000),under 10%PEG-6000 stress a significant reduction in germination potential(75.34%)and germination rate(42.38%)across were observed across 142 L.japonica germplasms,with notable variation in response magnitudes between these parameters.Similarly,exposure to 15%PEG-6000 led to an 8.24%decrease in root length and a 34.38%decline in shoot height,revealing distinct stress responses between root and shoot systems.These differential responses indicate substantial variability in drought resistance among the germplasms.To comprehensively evaluate drought resistance,we deployed correlation analysis,principal component analysis(PCA),membership function analysis,and cluster analysis.Based on this integrated assessment,the germplasm accessions were classified into six distinct drought-resistance categories:6 highly drought-resistant,9 drought-resistant,73 moderately resistant,41 drought-sensitive,and 13 highly sensitive.These findings provide a robust germplasm resource for the precise introduction and cultivation of L.japonica in both arid and non-arid regions,selective breeding of drought-tolerant varieties,and future investigations into the molecular regulatory networks underlying drought resistance in this species.
