摘要
野生大叶白麻广泛分布在塔里木河沿岸等地,经济价值高,被誉为“野生纤维素之王”。本研究为建立大叶白麻高效再生体系,促进定向选育优异品种,通过对大叶白麻种子采用不同的消毒方法进行处理,选择适宜的消毒方法;然后以大叶白麻无菌苗茎段为外植体,添加不同浓度的苯基噻二唑基脲(TDZ)、6-苄基氨基嘌呤(6-BA)、萘乙酸(NAA)等植物激素探讨对大叶白麻愈伤组织诱导和分化、不定芽生根的最佳激素配比。结果表明,较适宜的种子消毒方法为先用浓度为75%的乙醇消毒30 s,再用浓度为1%的过氧化氢消毒10 min,污染率为0.00%,萌发率为61.38%;MS+0.5 mg/L TDZ+0.2 mg/L NAA是适宜愈伤诱导和分化的培养基,诱导率和分化率分别达到90.67%和62.00%;最佳的不定芽生根培养基为1/2MS+0.5 mg/L NAA,生根率为100.00%。本研究成功建立了大叶白麻组织培养和植株再生体系,为大叶白麻良种繁育和遗传改良提供理论参考依据。
Poacynum hendersonii,which is widely distributed along the Tarim River,has a high value as natural fiber materials.This aim of this study was to establish an efficient callus regeneration system for rapid propagation and orientation breeding.In this study,the different disinfection methods were used to treat the seeds of P.hendersonii so as to select appropriate disinfection methods,and then the stem of P.hendersonii sterile seedlings were used as explants for rapid propagation.By adding different concentrations of plant hormones such as thidiazuron (TDZ), 6-Benzylaminopurine (6-BA), naphthylacetic acid (NAA), the optimal hormone ratio for the callus induction, the callus differentiation, and induction of adventitious roots of P. hendersonii was explored. The results showed that the most suitable sterilization method was as follows, first treated with 75% ethanol for 30 seconds, then treated with 1% hydrogen peroxide for 10 min, washed with sterile water 3-4 times, the pollution rate was 0. 00%, the germination rate was 61. 38%, and the germination was neat. The best medium for callus induction and differentiation was MS+0. 5 mg∕L TDZ+0. 2 mg∕L NAA, the induction rate was 90. 67%, and the induction rate of adventitious buds was 60. 00%;The best medium for rooting was 1∕2MS + 0. 5 mg∕L NAA, and the rooting rate could reach 100. 00%. This study provides technical support and theoretical reference for the rapid propagation of P. hendersonii, the production of secondary metabolites, and the research of molecular biology.
