摘要
以山莨菪种子为材料,采用单因素试验和正交试验研究不同消毒剂和消毒时间对山莨菪种子消毒和萌发的影响,探究种子的最佳消毒方法。结果表明,单独使用升汞或次氯酸钠都不能达到良好的消毒效果,升汞溶液对山莨菪种子的萌发具有抑制作用,次氯酸钠溶液的腐蚀作用则促进山莨菪种子的萌发;消毒剂组合进行正交试验得到山莨菪种子的最佳消毒方法:75%乙醇消毒0.5 min,再用75%次氯酸钠消毒15 min,最后用0.05%升汞消毒1.5 min,在此条件下山莨菪种子萌发率高达90.0%,染菌率为0,无菌苗生长快速。
Anisodus tanguticus seeds were used as materials,the single factor experiment and orthogonal experiment were used to study the effects of different disinfectants and disinfection time on the disinfection and germination of Anisodus tanguticus seeds,and the optimum disinfection method of the seeds was explored.The results showed that the use of mercuric chloride or sodium hypochlorite alone could not achieve good disinfection effect.The mercuric chloride solution inhibited the germination of Anisodus tanguticus seeds,while the corrosion of sodium hypochlorite solution promoted the germination of Anisodus tanguticus seeds.The optimum disinfection method of Anisodus tanguticus seeds was by orthogonal experiment of disinfectant combination:75%ethanol disinfection for 0.5 min,then 75%sodium hypochlorite disinfection for 15 min,and finally 0.05%mercuric chloride disinfection for 1.5 min.Under these conditions,the germination rate of Anisodus tanguticus seeds was as high as 90.0%,the bacterial infection rate was 0,and the aseptic seeding growed rapidly.
