摘要
为了解决野生薄皮木(Leptodermis oblonga Bge.)自然繁殖率低,提高种子萌芽率。本研究以野生薄皮木种子为试验材料,探究不同消毒剂种类、消毒时间等对野生薄皮木种子萌发的影响,优选出野生薄皮木组织培养中最佳的消毒灭菌方法。结果表明,相同消毒时间内,不同消毒剂对野生薄皮木种子萌芽率为:HgCl_(2)>NaClO>H_(2)O_(2)。三组消毒剂最佳处理时间为:酒精20 s、0.1%Hg Cl_(2)8 min种子萌芽率为53.09%、污染率为3.71%;酒精20 s、5%NaClO 10 min萌芽率为45.68%、污染率为29.63%;酒精30 s、10%H_(2)O_(2)10 min萌芽率为44.44%、污染率为14.81%通过野生薄皮木种子消毒剂组合的优化筛选,为野生薄皮木的快速繁殖技术和工厂化育苗提供理论依据和技术支持。
In order to solve the low reproduction rate of wild Leptodermis oblonga Bge.And improve the seed germination rate of wild Leptodermis oblonga Bge.In this study,we used the seeds of wild Leptodermis oblonga Bge.as experiment material,to explore the influence on the seed’s germination rate of wild Leptodermis oblonga Bge.,such as different disinfectant species,disinfection time,and then optimize the best disinfection and sterilization method in the tissue culture of wild Leptodermis oblonga Bge..The result shows that the seed germination rate of wild Leptodermis oblonga Bge.is HgCl_(2)>NaClO>H_(2)O_(2)in the same disinfection time.The best treatment time in the three combinations is:alcohol 20 s,0.1%Hg Cl_(2)8 min,seed germination rate is 53.09%,bacteria pollution rate is 3.71%;alcohol 20 s,5%NaClO 10 min germination rate is 45.68%,bacteria pollution rate is 29.63%;alcohol30 s,10%H_(2)O_(2)10 min germination rate is 44.44%,and bacteria pollution rate is 14.81%.It provides theoretical basis and technical support for the tissue culture and factory seedlings of wild Leptodermis oblonga Bge.from optimizing and screening the disinfection combination in wild Leptodermis oblonga Bge.seed.
