摘要
达氟沙星(DAN)是第三代氟喹诺酮类抗生素,因其具有广谱的抗菌活性而在畜牧业中广泛使用。DAN滥用会导致食品中抗生素残留,引发细菌耐药性等问题,从而对人类健康造成严重威胁。牛奶是一种常见的高营养食品,但奶牛容易富集DAN抗生素引发食品安全问题,因此对牛奶中DAN的快速灵敏检测非常重要。酶联免疫吸附法(ELISA)是一种基于抗原抗体特异性识别与酶催化信号放大的检测方法,具有高通量、低成本、快速等优点。本研究通过采用传统的ELISA实现了对牛奶中DAN的快速检测。实验结果表明,本方法检测DAN的标准曲线为y=0.999-0.258*[1-exp(-x/14.29)]-0.645*[1-exp(-x/1.16)],R^(2)=0.99,LOD值为0.89 ng/mL。本方法具有较高的灵敏度、特异性和准确性等优点,为食品中有害物质的检测提供了一种分析方法,为食品安全检测提供了技术支持。
As a third-generation fluoroquinolone antibiotic,danofloxacin(DAN)is widely used in the animal husbandry because of its broad-spectrum antibacterial activity.DAN abuse can lead to antibiotic residues in food,antimicrobial resistance and other problems,posing a serious threat to human health.Although the milk is a commonly-used high-nutrition food,DAN enrichment in dairy cows easily brings about some food safety problems.Therefore,it is of great significance to rapidly and sensitively detect DAN in milk.Enzyme linked immunosorbent assay(ELISA)is detection method with the advantages of high throughput,low cost and rapidity that combines antigen-antibody specificity recognition and enzyme-catalyzed signal amplification.This study ompleted the rapid detection of DAN in milk by the traditional ELISA.As is indicated by the experimental results,the standard curve of this method was y=0.999-0.258*[1-exp(-x/14.29)]-0.645*[1-exp(-x/1.16)],R^(2)=0.99,with a limit of detection(LOD)of 0.89 ng/mL.The method is featured with high sensitivity,specificity and accuracy,which provides an analytical method for detecting harmful substances in food and technical support for food safety detection.
