摘要
为建立一种动物源性食品(鸡肉、猪肉以及鸡蛋)中8种氟喹诺酮类药物(氧氟沙星、诺氟沙星、培氟沙星、环丙沙星、洛美沙星、达氟沙星、恩诺沙星、沙拉沙星)多残留的高效液相色谱检测方法,将鸡肉、猪肉以及鸡蛋样品绞碎或匀浆后经磷酸盐缓冲溶液提取(鸡蛋样品需增加正己烷脱脂步骤),以Waters Oasis HLB固相萃取柱净化,用高效液相色谱法进行检测。结果显示:在最优试验条件下测得8种氟喹诺酮类药物线性关系良好,相关系数(R^(2))均大于0.999;本方法对8种药物的检出限为0.4~2.0μg/kg,定量限为2.0~10.0μg/kg;在3个残留量添加水平下,8种药物的平均回收率为83.3%~110.0%,相对标准偏差(RSD)为1.1%~4.8%。结果表明,该方法准确、灵敏、稳定,达到了兽药残留检测要求,可为基层动物源性食品中限用和停用氟喹诺酮类药物的多残留检测提供技术参考。
In order to establish a high performance liquid chromatography(HPLC)for multiple residues of 8 fluoroquinolones(ofloxacin,norfloxacin,pefloxacin,ciprofloxacin,lomefloxacin,dafloxacin,enrofloxacin and salafloxaci)in animal-derived food(chicken,pork and eggs),the samples of chicken,pork and eggs were minced or homogenized and extracted with phosphate buffer solution(N-hexane degreasing needed for eggs),purified with Waters Oasis HLB solid phase extraction column,and detected by HPLC.The results showed that the linearities of the 8 fluoroquinolones were good under the optimal test conditions,and the correlation coefficients(R^(2))were all larger than 0.999;the detection limits ranged from 0.4 to 2μg/kg,and the quantitation limits were from 2 to 10μg/kg;the average recoveries of the 8 drugs were from 83.3%to 110.0%at the addition levels of 3 residues,and the relative standard deviations(RSD)were from 1.1%to 4.8%.In conclusion,the method was accurate,sensitive and stable,and capable of detecting veterinary drug residues,which provided a technical reference for detecting multiple residues of fluoroquinolones restricted or banned in animal-derived food at grassroots level.
