摘要
                
                    目的建立橘红的高效液相色谱法(HPLC)特征图谱和绿原酸、芸香柚皮苷、橙皮苷、川陈皮素、橘皮素5个成分含量测定方法,为橘红饮片的质量控制提供参考。方法色谱柱Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5μm);流动相:乙腈(A)-0.1%甲酸溶液(B),梯度洗脱;流速1.0 mL/min;检测波长280 nm;色谱柱温度30℃;进样量10μL。采集10批橘红样品色谱数据,建立橘红的特征图谱。对绿原酸、芸香柚皮苷、橙皮苷、川陈皮素和橘皮素的含量测定方法进行方法学考察,并测定上述5个成分含量。结果10批橘红样品的特征图谱含有25个共有峰,相似度为0.910~0.998,指认出绿原酸、芸香柚皮苷、橙皮苷、川陈皮素、橘皮素、芦丁6个成分。方法学考察结果显示:绿原酸、芸香柚皮苷、橙皮苷、川陈皮素、橘皮素在0.0095~0.0424、0.0246~0.1096、0.0926~0.4136、0.0179~0.0800、0.0095~0.0424 mg/mL范围内线性关系良好,r均>0.9990。10批样品中绿原酸、芸香柚皮苷、橙皮苷、川陈皮素和橘皮素的含量范围分别为0.5134~0.6096、4.7224~4.9921、42.3494~45.9137、0.6024~0.6536、0.3070~0.3337 mg/g,其平均加样回收率为97.79%~104.40%,RSD为1.18%~6.29%。结论本实验建立了橘红的HPLC特征图谱和5个成分的含量测定方法,具有较高的稳定性和可行性,可为橘红的质量控制提供参考。
                
                Objective:To establish the high performance liquid chromatographic(HPLC)characteristic spectrum of Citri Eoxcarpium Rubrum and the determination method for five components including chlorogenic acid,rutin naringin,hesperidin,nobiletin,tangeretin,which can provide a reference for the quality control of Citri Eoxcarpium Rubrum.Methods:Agilent ZORBAX SB-C_(18)(4.6 mm×250 mm,5μm)was used as the chromatographic column;the mobile phase was acetonitrile(A)-0.1%formic acid solution(B)with gradient elution;the flow rate was 1.0 mL/min;the detection wavelength was 280 nm;the column temperature was 30℃;the injection volume was 10μL.10 batches of Citri Eoxcarpium Rubrum chromatographic data were collected,and the characteristic spectrum was established.Methodological investigation was conducted on the determination methods of chlorogenic acid,rutin naringin,hesperidin,nobiletin and tangeretin,and the contents of the five components were determined.Results:The characteristic spectrum of 10 batches of Citri Eoxcarpium Rubrum samples contained 25 shared peaks with similarities ranging from 0.910~0.998.Six components including chlorogenic acid,rutin naringin,hesperidin,nobiletin,tangeretin and rutin were identified.The methodological investigation results showed that chlorogenic acid,rutin naringin,hesperidin,nobiletin,and tangeretin had good linear relationships within the ranges of 0.0095-0.0424,0.0246-0.1096,0.0926-0.4136,0.0179-0.0800,0.0095-0.0424 mg/mL,with r values greater than 0.9990.The content ranges of chlorogenic acid,rutin naringin,hesperidin,nobiletin,and tangeretin in 10 batches of samples were 0.5134-0.6096,4.7224-4.9921,42.3494-45.9137,0.6024-0.6536,0.3070-0.3337 mg/g respectively.The average recovery rate was 97.79%to 104.40%,and the range of RSD was 1.18%to 6.29%.Conclusion:This experiment established an HPLC characteristic spectrum and content determination method for five components of Citri Eoxcarpium Rubrum,which has high stability and feasibility,and can provide reference for the quality control of Citri Eoxcarpium Rubrum.
