青皮及其炮制品UPLC指纹图谱与多成分含量测定研究 被引量:7

Study on UPLC Fingerprint and Multi-Components Content Determination of Citri Reticulatae Pericarpium Viride and Its Processed Products

摘要 目的:建立青皮及醋炙品UPLC指纹图谱方法,对其中4个成分进行含量测定,为其不同规格及相应炮制品的质量标准建立提供依据。方法:采用ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)色谱柱,以乙腈-0.1%甲酸溶液为流动相,梯度洗脱,建立了52批不同规格青皮及其炮制品的指纹图谱,并测定4个成分含量,运用相关软件进行数据分析。结果:青皮及炮制品UPLC特征图谱中确定11个共有峰,正交偏最小二乘判别分析可区分个青皮与四花青皮(包括其炮制品),生品与炮制品间区分不明显;同时对已归属的4个成分进行含量测定,运用SPSS 20.0软件进行结果分析,发现辛弗林和橙皮苷的含量在不同规格青皮间(包括其对应炮制品)存在差异,而辛弗林的含量在四花青皮炮制前后变化较大。结论:该研究建立的UPLC指纹图谱和多成分含量测定方法稳定,专属性强,可为青皮及其炮制品的质量评价提供参考。 Objective: To establish the UPLC fingerprint of Citri Reticulatae Pericarpium Viride and its processed products fried with vinegar and determine the content of four components, so as to provide a basis for the establishment of quality standards for different specifications and corresponding processed products.Methods: The chromatographic column was ACQUITY UPLC HSS T3(100 mm×2.1 mm, 1.8 μm) column, the acetonitrile-0.1%formic acid solution was used as the mobile phase for gradient elution to establish the fingerprints of 52 batches of different specifications of Citri Reticulatae Pericarpium Viride and its processed products, the contents of four components were determined and the relevant software was used for data analysis.Results: There were 11 common peaks in the UPLC fingerprints of Citri Reticulatae Pericarpium Viride and its processed products.Orthogonal partial least squares discriminant analysis could distinguish Citri Reticulatae Pericarpium Viride "Geqingpi" and Citri Reticulatae Pericarpium Viride "Sihuaqingpi",including the processed products, there was no significant difference between raw products and processed products.At the same time, the contents of the four components were determined, and the results were analyzed by SPSS 20.0 software.It was found that the contents of synephrine and hesperidin were different among different specifications of Citri Reticulatae Pericarpium Viride, including corresponding processed products, and the content of synephrine changed greatly in Citri Reticulatae Pericarpium Viride "Sihuaqingpi" before and after processing.Conclusion: The UPLC fingerprint and multi-component content determination method established in this study is stable and specific, which can provide reference for the quality evaluation of Citri Reticulatae Pericarpium Viride and its processed products.
