摘要
为应对果蔬中多种特定农药残留快速检测的需求,本文基于胶体金免疫层析技术,通过优化胶体金-单克隆抗体偶联物浓度、人工抗原包被浓度等关键因素,研制出克百威、灭多威、丙溴磷等15种农药残留同步快速检测的胶体金免疫层析多联卡。测试结果显示,15种农药残留的检出限在0.01~0.3 mg·kg^(-1),未发现假阴性情况,假阳性率均<5%,特异性和准确性均>95%。实际应用中共检测2516批次样品,检出阳性样品6批次。该方法准确性好、操作简单,适用于蔬果多农药残留的高通量快速筛查检测。
In order to meet the needs of rapid detection of a variety of specific pesticide residues in fruits and vegetables,based on colloidal gold immunochromatography technology,this paper developed a colloidal gold immunochromatographic multicard for the simultaneous rapid detection of 15 pesticide residues,such as carbofuran,methocarb and profenofos,by optimizing the key factors such as colloidal gold-monoclonal antibody conjugate concentration and artificial antigen coating concentration.The test results showed that the detection limit of 15 pesticide residues was 0.01 mg·kg^(-1) to 0.3 mg·kg^(-1),no false negatives were found,the false positive rate was less than 5%,and the specificity and accuracy were greater than 95%.A total of 2516 batches of samples were tested,and 6 batches of positive samples were detected.This method has good accuracy and simple operation,and is suitable for high-throughput rapid screening and detection of pesticide residues in vegetables and fruits.
