红景天UGTs基因与有效成分含量的相关性研究 被引量:5

Correlation Analysis between UGTs Gene Sequence and Effective Composition of Salidrosides in Different Rhodiola Species

摘要 本研究采集青藏高原地区不同种的红景天植株,采用改良CTAB法提取红景天总DNA。PCR扩增UGTs基因片段,对扩增产物进行序列测定,分析红景天UGTs基因多态性。同时,采用高效液相色谱法(HPLC)测定不同植株中红景天苷、酪醇、没食子酸的含量,分析三种有效成分的含量差异。结果表明:不同红景天植物UGTs基因序列存在多样性,三种有效成分的含量也存在一定差异。7个样品为7种不同的单倍型,不同种的红景天中有效成分的含量与地理分布有一定的联系,不同植株不同单倍型和同一植株不同单倍型所含的红景天苷、酪醇和没食子酸的含量都存在一定的差异,选用的红景天植物材料所对应的单倍型与红景天苷含量、酪醇含量之间无显著性差异,与没食子酸含量之间差异显著。 We collected different species Rhodiola plants in Tibet Plateau, extracted the total DNA by modified CTAB method. The gene polymorphisdm of Rhodiola was analyzed, after UGTs gene sequences were amplified by PCR, and the products were sequenced. At the same time, the content of effective compositions salidroside, tyrosol and gallic acid in different species of Rhodiola plants was determined by HPLC. The results showed that the genetic diversity of different species Rhodiola plants was verified and there are differences in the content of effective compositions. The UGTs sequences of the seven Rhodiola samples can be divided into seven genotypes and the content of effective compositions is relevant with the geographical distribution. Salidroside, tyrosol and gallic acid in different plants and different haploid types of the same plants have a certain degree of differences.There was no significant difference between Salidroside and tyrosol in the corresponding haplotype plants, but significant difference between gallic acid.
