蒲公英水提取物对副溶血弧菌的抗菌活性

摘要 通过测定最小抑菌浓度(minimal inhibitory concentration,MIC)、生长曲线、浮游细胞失活率评价蒲公英水提取物对副溶血弧菌的抑菌活性,并通过测定胞外碱性磷酸酶与蛋白质泄漏、菌体运动能力以及菌体生物膜形成抑制率探究其抗菌机理。结果表明:蒲公英水提取物对副溶血弧菌的MIC为26 mg/mL,能显著抑制其生长和繁殖。在生理盐水条件下,1 MIC的蒲公英水提取物作用于菌体3 h可使其失活率达到99.04%,且菌体胞外碱性磷酸酶活性和蛋白含量分别提高了4.34倍和5.61倍。扫描电镜结果显示蒲公英水提取物改变了菌体细胞形态,从而有效抑制副溶血弧菌游动和群集的能力。此外,1/4 MIC和1/2 MIC蒲公英水提取物对副溶血弧菌生物膜形成抑制率分别48.59%和71.31%。因此,蒲公英水提取物具有潜在应用价值,可用于水产饲料或水产养殖水体中控制副溶血弧菌。 The aim of this study was to investigate the antibacterial activity and mechanism of action of natural antimicrobial agent dandelion water extract against Vibrio parahaemolyticus.The antibacterial activity was evaluated by determining the minimal inhibitory concentration(MIC),growth curve,and planktonic cell inactivation rate.The antibacterial mechanism was explored by assessing extracellular al⁃kaline phosphatase and protein leakage,bacterial motility,and inhibition rate of biofilm formation.The results showed that the MIC of Dandelion water extract against V.parahaemolyticus was 26 mg/mL,sig⁃nificantly inhibiting its growth and proliferation.Under physiological saline conditions,treatment with 1 MIC of dandelion water extract for 3 hours resulted in a 99.04%inactivation rate,with extracellular al⁃kaline phosphatase activity and protein content increasing by 4.34-fold and 5.61-fold,respectively.Scan⁃ning electron microscopy revealed changes in bacterial cell morphology induced by Dandelion water ex⁃tract,effectively inhibiting V.parahaemolyticus swimming and swarming ability.Furthermore,dandelion water extract at 1/4 MIC and 1/2 MIC inhibited biofilm formation of V.parahaemolyticus by 48.59%and 71.31%,respectively.Therefore,Dandelion water extract holds potential for application in control⁃ling V.parahaemolyticus in aquaculture feed or water.
