摘要
【目的】鉴定玉米Rpp13(Recognition of Peronospora parasitica 13)基因家族成员是否与玉米穗腐病(ear rot)抗性相关,为丰富玉米穗腐病抗病基因资源奠定基础。【方法】采用关联分析、转录组分析、实时荧光定量PCR(RT-qPCR)、候选基因分析及单倍型分析的方法,对玉米抗穗腐病基因进行鉴定及功能分析。【结果】从玉米全基因组中鉴定出56个玉米Rpp13基因,根据物理位置的先后顺序命名为ZmRpp13-1~ZmRpp13-56。对玉米Rpp13基因自然变异位点与穗腐病抗性进行候选基因关联分析,挖掘出玉米Rpp13基因家族重要的抗穗腐病候选基因,结合抗性转录组分析,表明了Zm00001d037650(ZmRpp13-34)与玉米穗腐病抗性极显著相关。使用RT-qPCR的方法鉴定出ZmRpp13-34在拟轮枝镰孢接种后的基因表达水平上调。对ZmRpp13-34进行单倍型分析确定了影响穗腐病抗性的功能单倍型。【结论】玉米Rpp13遗传变异与穗腐病抗性显著相关,其中ZmRpp13-34可作为调控玉米穗腐病抗性的重要候选基因。
【Objective】This study aims to identify the members of maize Rpp13(Recognition of Peronospora parasitica 13)gene family associated with resistance to maize ear rot resistance,thereby laying a foundation for enriching maize ear rot resistance gene resources.【Method】Through association analysis,transcriptome analysis,real-time fluorescence quantitative PCR(RT-qPCR),candidate gene analysis and haplotype analysis,the identification and functional analysis of maize ear rot resistance genes were carried out.【Result】Fifty-six maize Rpp13 genes were identified from the maize genome and named as ZmRpp13-1 through ZmRpp13-56 according to their physical order.Through candidate gene association analysis between natural variation sites in maize Rpp13 genes and resistance to ear rot disease,several significant candidate genes conferring resistance to ear rot were discovered within the maize Rpp13 gene family.Zm00001d037650(ZmRpp13-34)showed the most pronounced association with resistance to maize ear rot based on RNA sequencing and analysis.RT-qPCR revealed that the relative expression level of ZmRpp13-34 was significantly upregulated post-inoculation with Fusarium verticillioides.The Haplotype analysis of ZmRpp13-34 pinpointed functional haplotypes affecting resistance to ear rot.【Conclusion】The results showed that the genetic variation of maize Rpp13 gene was significantly correlated with ear rot resistance,and ZmRpp13-34 emerges as a critical candidate gene involved in regulating maize resistance to ear rot.
