基于指纹图谱及多化学成分分析的生半夏及其炮制品质量研究 被引量:1

Quality Evaluation of Pinelliae Rhizoma and Processed Products Based on HPLC Fingerprint and Multi-content Determination

摘要 目的评价各产区半夏及其炮制品的质量,分析半夏经硫熏后水溶性成分含量的变化。方法采用HPLC法建立39批次半夏及其炮制品水溶性核苷类成分化学指纹图谱方法,标定了16个共有峰,指认了鸟苷、腺苷、次黄嘌呤、尿苷等9种核苷类成分。采用中药相似度评价系统(2012年版),对半夏及其炮制品水溶性核苷类成分进行相似度评价;采用SIMCA 14.1对共有成分的相对含量进行降维,通过正交偏最小二乘法(OPLS-DA)有效识别南通地产野生半夏和全国范围内半夏资源的质量差异,对不同半夏炮制品进行区别。建立鸟苷等9种核苷类成分的含量测定方法,考察硫熏加工对核苷类成分的影响。结果以南通地产野生半夏为参照,其他半夏及炮制品的相似度在0.635~0.977,所建立的指纹图谱方法可用于半夏生品与炮制品的整体质量控制,指纹图谱的相似度结果及OPLS-DA均能有效识别半夏生品及炮制品,通过OPLS-DA得分图将半夏的不同炮制品也进行了有效的分类。相似度评价系统和主成分分析均未体现硫熏对半夏核苷类成分的影响。结论所建立的方法简便、准确,可实现基于HPLC指纹图谱和多化学成分分析的半夏质量研究。硫熏处理对半夏核苷类成分未产生显著影响,合理的硫熏仍可认为是当前半夏快速干燥的有效途径。该结果可为半夏质量标准的改进和建立提供数据支撑。 Objective To evaluate the quality of pinelliae rhizoma and processed products in various production areas and analyze the changes in the content of water-soluble components after sulfur fumigation.Methods Establish a chemical HPLC fingerprint of water-soluble nucleosides in 39 batches of pinelliae rhizoma and processed products.Sixteen common peaks were calibrated,and nine nucleosides,including guanosine,adenosine,hypoxanthine and uridine were identified.SIMCA 14.1 reduced the relative content of common components and effectively identified the quality differences between wild pinelliae rhizoma in Nantong and nationwide resources through OPLS-DA,distinguishing the processed products.Establish a method for determining the content of nine nucleoside components such as guanosine,and investigate the effect of sulfur fumigation on nucleoside components.Results Taking wild pinelliae rhizoma from Nantong as a reference,the similarity ranges of the other pinelliae rhizoma and processed products were 0.635 to 0.977.The established fingerprint method can be used for overall quality control of pinelliae rhizoma and processed products,and the similarity results of the fingerprint can effectively identify pinelliae rhizoma and processed products.Pinelliae rhizoma and processed products were effectively classified by orthogonal partial least squares discriminate analysis(OPLS-DA),also the different processed products.Neither the similarity evaluation system nor the principal component analysis showed the influence of sulfur fumigation on the nucleosides of pinelliae rhizoma and processed products.Conclusion The established method is simple and accurate,and can be used to study the quality of pinelliae rhizoma based on HPLC fingerprint and multi chemical component analysis.Sulfur fumigation has no significant effect on the nucleoside components of pinelliae rhizoma.Reasonable sulfur fumigation can still be considered as an effective way for rapid drying of pinelliae rhizoma.The results can provide data support for the improvement and establishment of quality standards for pinelliae rhizoma.
