摘要
代谢组学利用现代分析技术分析生物体内内源性代谢物的变化,结合生物信息学方法探索生物标记物和代谢途径,揭示病理机制和药物干预机制。代谢组学强调机体整体状态变化与中医药的整体观诊疗思维相匹配,两者有机结合在复杂的中医药理论研究中具有广阔的应用前景。液质联用(LCMS)技术具有分离性能好、灵敏度高和专属性强等优势,是最适用于中药代谢组学的定性定量分析技术。通过总结LC-MS的常用类型和生物样品的前处理方法,以及近3年LC-MS在中药代谢组学中的具体应用,以期为中药代谢组学研究提供借鉴。
Metabonomics uses modern analytical techniques to analyze the changes of endogenous metabolites in organisms,and explores biomarkers and metabolic pathways in combination with bioinformatics methods to reveal the pathological mechanism and drug intervention mechanism.Metabonomics emphasizes that the change of the whole state of the body is matched with the thinking of diagnosis and treatment of the holistic view of Traditional Chinese Medicine(TCM).The organic combination of the two has broad prospects in the complex theoretical research of TCM.liquid chromatography-mass spectrometry(LC-MS)has the advantages of good separation performance,high sensitivity and strong specificity.It is the most suitable qualitative and quantitative analysis technology for metabonomics of TCM.This paper summarized the common types of LC-MS and the pretreatment methods of biological samples,as well as the specific application of LC-MS in metabonomics of TCM in recent three years,in order to provide reference for metabonomics research of TCM.
