摘要
目的:研究尾静脉注射和灌胃维生素B_(6)对盐酸多奈哌齐大鼠体内药动学的影响。方法:18只雄性SD大鼠,随机数字表法均分为盐酸多奈哌齐组、尾静脉注射维生素B_(6)联合给药组和灌胃维生素B_(6)联合给药组。使用超高效液相色谱-荧光法测定血药浓度并绘制药-时曲线,DAS 2.0软件拟合药动学参数,使用SPSS 26.0软件对组间药动学参数进行统计学分析。结果:与盐酸多奈哌齐组相比,尾静脉注射维生素B_(6)联合给药组MRT_(0-t)、MRT_(0-∞)、t_(1/2z)均显著延长(P<0.01),V_(z/F)显著增大(P<0.01),C_(max)显著降低(P<0.01);灌胃维生素B_(6)联合给药组AUC_(0-t)、AUC_(0-∞)、C_(max)均显著降低(P<0.01),MRT_(0-t)、MRT_(0-∞)、t_(1/2z)显著延长(P<0.01,P<0.05),CL_(z/F)、V_(z/F)显著增大(P<0.01)。结论:尾静脉注射和灌胃维生素B_(6)均可降低大鼠体内盐酸多奈哌齐的血药浓度,减缓盐酸多奈哌齐在大鼠体内的代谢,延长其在大鼠体内的作用时间。
OBJECTIVE To study the effect of tail vein injection and gavage administration of vitamin B_(6)on the pharmacoki⁃netics of donepezil hydrochloride in rats.METHODS Totally 18 male SD rats were randomly and equally divided into donepezil hydrochloride group,tail vein vitamin B_(6)co-administration group and gavage vitamin B_(6)co-administration group.The blood con⁃centration of drug was determined by ultra-high performance liquid chomatography-fluorescence method,and the drug concentration-time curve was drawn.The pharmacokinetic parameters were fitted with DAS 2.0 software and statistically ana⁃lyzed using SPSS 26.0 software.RESULTS Compared with donepezil hydrochloride group,the tail vein vitamin B_(6)coadministration group had significantly longer MRT_(0-t),MRT_(0-∞)and t_(1/2z)(P<0.01),significantly greater V_(z/F)(P<0.01),and sig⁃nificantly lower C_(max)(P<0.01);the gavage vitamin B_(6)co-administration group had significantly lower AUC_(0-t),AUC_(0-∞),and C_(max)(P<0.01),MRT_(0-t),MRT_(0-∞)and t_(1/2z)were significantly prolonged(P<0.01,P<0.05),and CLz/F and Vz/F were significantly increased(P<0.01).CONCLUSION Both tail vein injection and intragastric administration of vitamin B_(6)can reduce the plasma concentration of donepezil hydrochloride,slow down the metabolism of donepezil hydrochloride,and prolong its action time in rats.
