摘要
建立超高效液相色谱-串联质谱法快速筛查化妆品中40种抗感染药物的方法。样品采用含体积分数0.5%甲酸的乙腈溶液超声提取,经Acquity UPLC BEH C18色谱柱(100 mm×2.1 mm×1.7μm)分离,以5 mmol/L乙酸铵缓冲溶液(p H=4.0)和体积分数0.1%甲酸-乙腈为流动相进行梯度洗脱,流速为0.3 mL/min,柱温为40℃,进样量为1μL,采用电喷雾离子源,正/负离子多反应监测模式,外标法定量。这40种抗感染药物在25~1 000 ng/mL范围内线性关系良好,相关系数均大于0.99;相对标准偏差为0.57%~8.85%;回收率为82.25%~117.98%;检出限为0.01~1.75 ng/mL。该方法操作简便,实用性强,分析速度快,可用于化妆品中抗感染类药物的快速筛查。
An analytical method was established to fast screen 40 kinds of anti-infective drugs in cosmetics by ultra performance liquid chromatograph-tandem mass spectrometry(UPLC-MS/MS)method.The samples were extracted by ultrasonic extraction with acetonitrile containing 0.5%(volume fraction)formic acid,and separated on Acquity UPLC BEH C18 column(100 mm×2.1 mm×1.7μm)with gradient elution program using 5 mmol/L buffer solution of ammonium acetate(pH=4.0)and 0.1%(volume fraction)formic acid-acetonitrile at the flow rate of 0.3 mL/min and the column temperature of 40℃.The injection volume was 1μL.The samples were detected by electrospray ion source with positive/negative ion multi-reaction monitoring mode(MRM),and quantified through the external standard.The linear relationship of the 40 kinds of anti-infective drugs was good within the concentration range of 25-1000 ng/mL,and the correlation coefficients were greater than 0.99.The relative standard deviations were 0.57%-8.85%,the recoveries were within 82.25%-117.98%,and the limits of detection were 0.01-1.75 ng/mL.The method is simple,practical and fast,which can be used for rapid screening of anti-infective drugs in cosmetics.
