响应面法优化超声提取益母草中多酚工艺及其抗氧化作用研究 被引量:1

Polyphenols from Leonurus japonicus Houtt:Optimization of Ultrasonic Extraction Conditions by Response Surface Methodology and Antioxidant Activity

摘要 目的:优选出益母草中多酚提取工艺,并对其进行抗氧化活性研究。方法:在单因素实验基础上,以没食子酸标准品为对照,以乙醇浓度、料液比、超声时间为自变量,以益母草多酚得率为响应值,运用响应面法优化超声辅助提取工艺参数;并通过DPPH、ABTS自由基法研究益母草多酚体外抗氧化作用。结果:益母草中多酚最优工艺为:乙醇浓度42%,料液比1∶8(g/mL),超声时间15 min,此时得率为3.78%,与理论得率3.82%,相差0.04%,工艺稳定、可行,可用于益母草中多酚提取。抗氧化实验可知,益母草多酚和维生素C对DPPH、ABTS的清除率均随二者在测试浓度(4~14 mg/mL)范围内正相关;当浓度为14 mg/mL时,维生素C和益母草多酚对DPPH的平均清除率分别是89.34%和75.96%,差异不明显;对ABTS的平均清除率分别是79.52%和61.81%,差异在高浓度时较为显著。结论:益母草多酚具有一定的抗氧化作用。该研究为益母草资源广泛开发提供参考。 Objective:To optimize the extraction process of polyphenols from Leonurus japonicus Houtt and study their antioxidant activity.Methods:On the basis of single factor experiments,the extraction conditions including ethanol concentration,solid-to-liquid ratio,and ultrasonic time were optimized by response surface methodology with the yield of polyphenols as the response value and gallic acid standard as the control.Furthermore,the antioxidant activity of the obtained polyphenols in vitro was evaluated based on the DPPH and ABTS free radical scavenging activities.Results:The optimum extraction conditions of polyphenols from L.japonicus were as follows:ultrasonic extraction with 42% ethanol and the solid-to-liquid ratio of 1∶8(g/mL) for 15 min,under which the yield was 3.78%,with a difference of 0.04% compared with the theoretical yield of 3.82%.The process was stable and feasible and can be used for the extraction of polyphenols from L.japonicus.The scavenging rates of L.japonicas polyphenols and VC against DPPH and ABTS free radicals were positively correlated with the concentration within the range of 4 to 14 mg/mL.The average scavenging rates of 14 mg/mL VC and L.japonicas polyphenols against DPPH free radicals were 89.34% and 75.96%,respectively,without significant difference.The average scavenging rates of VC and L.japonicas polyphenols against ABTS free radicals were 79.52% and 61.81%,respectively,which showed large differences at high concentrations.Conclusion:L.japonicas polyphenols had antioxidant activity.This study provides a reference for mining L.japonicus resources.
