甘露消毒丹中芳香药物防治流感的作用机制探讨——基于网络药理学 被引量:5

摘要 目的:运用网络药理学方法分析探讨甘露消毒丹中芳香药物防治流感的作用机制。方法:采用TCMSP数据库及分析平台获取甘露消毒丹中芳香药物的成分信息;通过GeneCards数据库获取流感的疾病靶点信息,并对甘露消毒丹中的芳香药物靶点进行维恩分析,获得交集靶点。利用STRING网站分析靶点间的相互作用,运用Cytoscape3.7.2软件进行网络分析并筛选关键靶点,应用Metascape数据库对关键靶点进行GO与KEGG富集分析。结果:甘露消毒丹的芳香药物治疗流感的关键靶点共24个,其中,主要的靶点包括IL-6、IL-8、TNF、IL-10、IL1B、CCL2、CXCL8、EGFR、STAT1、AP-1、JUN、Caspase-3等。经GO分析共获得相关的生物学过程967条,细胞结构28个,分子功能46条,主要涉及对氧化应激的反应、对活性氧的反应、对无机物质的反应、对脂多糖的反应、对细菌来源分子的反应等生物学过程,膜筏、膜微区、膜区、囊泡腔、小窝、质膜筏等细胞结构,以及细胞因子受体结合、细胞因子活性、受体调节剂活性、核心启动子结合、肿瘤坏死因子受体超家族结合等分子功能。KEGG富集分析共获得相关通路114条。其中,主要的通路包括IL-17信号通路、TNF信号通路、Toll样受体信号通路、NOD样受体信号通路、MAPK信号通路、癌症通路等。结论:甘露消毒丹中的芳香药物主要通过激活机体适度的免疫炎症反应,并抑制过度的炎症反应,从而发挥其抗病毒与抗炎的作用。 Objective:Using network pharmacology method to analyze and explore the mechanism of aromatic drugs in Ganlu Xiaodu Decoction in preventing and treating influenza.Methods:The TCMSP Database and Analysis Platform were used to obtain the component information of aromatic drugs in Ganlu Xiaodu Decoction.Influenza disease target information was obtained from the GeneCards database,and Venny analysis was performed with aromatic drug targets in Ganlu Xiaodu Decoction to obtain intersecting targets.The interaction between the targets was analyzed by STRING website,the network analysis was carried out by using the software of Cytoscape 3.7.2,and the enrichment analysis of GO and KEGG was carried out based on the Metascape database.Results:A total of 24 key targets for the aromatic drug treatment of influenza were identified.The main targets include IL-6,IL-8,TNF,IL-10,IL1 B,CCL2,CXCL8,EGFR,STAT1,AP-1,JUN,Caspase-3,etc.A total of 967 relevant biological processes,28 cell structures and 46 molecular functions were obtained by GO analysis,mainly involving biological processes such as response to oxidative stress,reaction to reactive oxygen species,reaction to inorganic substances,reaction to lipopolysaccharides,reaction to bacterial-derived molecules and cell structures such as membrane rafts,membrane microdomains,membrane regions,vesicle cavities,caveolae,plasma membrane rafts,and molecular functions such as cytokine receptor binding,cytokine activity,receptor regulator activity,core promoter binding,tumor necrosis factor receptor superfamily binding,etc.A total of 114 related pathways were obtained by KEGG enrichment analysis.The main pathways include IL-17 signaling pathway,TNF signaling pathway,Toll-like receptor signaling pathway,NOD-like receptor signaling pathway,MAPK signaling pathway,cancer pathway,etc.Conclusion:The aromatic drugs in Ganlu Xiaodu Decoction play the role of antiviral and anti-inflammatory mainly by activating the moderate immune inflammatory reaction and inhibiting the excessive inflammatory reaction.
