摘要
文章以紫苏叶为研究对象,采用响应面分析法优化紫苏叶总黄酮的提取工艺。通过醇提法对紫苏叶总黄酮进行提取,首先对提取时间、提取温度、乙醇浓度和料液比4个单因素条件进行研究,然后进行响应面试验设计,优化分析紫苏叶总黄酮的提取工艺。结果表明,此方法提取紫苏叶中总黄酮的最佳工艺为:提取时间4.5 h,乙醇浓度68%,提取温度65℃,料液比1:55(g/mL),其理论提取率为7.48%,实际提取率为7.43%,提取温度、乙醇浓度、料液比、提取时间4个单因素对提取率的影响程度由大到小。验证试验结果表明,其结果与响应面分析预测的结果一致,该模型可靠,该研究为提高紫苏叶总黄酮提取率提供了参考。
In this paper,response surface analysis was used to optimize the extraction process of total flavonoids from Perilla leaves.The extraction of total flavonoids from perilla leaves was carried out by alcohol extraction method.First,four single factors such as extraction time,extraction temperature,ethanol concentration and solid-liquid ratio were investigated.Then the response surface experiment was designed to optimize the extraction process of total flavonoids from Perilla leaves.The results showed that the optimal extraction conditions were as follows:extraction time 4.5 h,ethanol concentration 68%,extraction temperature 65℃,solid-liquid ratio 1:55(g/mL),the theoretical extraction rate was 7.48%,the actual extraction rate was 7.43%,four single factors,extraction temperature,ethanol concentration,solid-liquid ratio and extraction time,had great influence on the extraction rate from large to small.The experimental results show that the model is reliable and consistent with the predicted results of response surface analysis.This study provides a reference for improving the extraction rate of total flavonoids from Perilla leaves.
