摘要
本文以万寿菊为研究对象,采用超声提取万寿菊不同部位(花、茎、根)中的多酚、黄酮类物质并进行含量测定,并研究它们的抗氧化活性。利用单因素实验分别对提取溶剂、溶剂浓度、料液比、提取时间等影响因素对万寿菊茎中多酚和黄酮含量进行评价。结果显示:80%甲醇溶液作为提取溶剂,料液比1∶80(g/mL),超声提取周期40 min为最佳提取工艺。通过该工艺测得万寿菊花、茎、根各部位中多酚含量分别为15.34、2.04、1.03 mg/g,黄酮含量分别为44.10、2.69、3.38 mg/g,DPPH自由基清除能力的抗氧化活性的IC50分别是1.03±0.03 ng/L、68.5±5.01 mg/mL、98.90±18.91 mg/mL,抗氧化活性:花>茎>根;ABTS+自由基清除能力的抗氧化活性分别为1.34±0.02 ng/L、139.4±9.01 mg/mL、92.30±16.80 mg/mL,抗氧化活性:花>根>茎。进一步相关性统计分析表明,万寿菊不同部位提取物中多酚含量对DPPH和ABTS自由基清除能力表现出显著的正相关,而黄酮含量与其抗氧化活性相关性不显著。
Tagetes erecta L.(marigold)was taken as the study object,flavonoids and polyphenols were extracted from different parts of marigold(flower,stem and root)by ultrasonic extraction,the contents were determined and the antioxidant activity was studied.The optimal extraction procedure was evaluated by the following four contributing factors:extraction solvents,extraction solvent concentration,extraction time and material-to-liquid ratio with single factor experiments.The results showed that the optimal extraction process was as follow:80%methanol,ultrasonic extraction for 40 min and solid-liquid ratio of 1:80 g/mL.The flavonoids and polyphenols in marigold flowers,stems and roots were extracted with this process,and the content of polyphenols was determined to be 15.34,2.04,1.03 mg/g respectively,and the content of flavonoids were 44.1,2.69,3.38 mg/g,respectively.The IC50 of the antioxidant activity of DPPH free radical scavenging ability was 1.03±0.03 ng/L,68.5±5.01 mg/mL,98.90±18.91 mg/mL respectively,which represented the antioxidant activity order was flower>stem>root;The antioxidant activity of ABTS free radical scavenging ability was 1.34±0.02 ng/L,139.4±9.01 mg/mL,92.30±16.80 mg/mL respectively,and the antioxidant activity order was flower>root>stem.Correlation analysis found that DPPH and ABTS free radical scavenging ability of different parts of marigold extracts and polyphenol contents showed a significant positive correlation,there was no significant correlation between flavonoid content and antioxidant activity.
