防风中5-O-甲基维斯阿米醇苷抗炎作用及机制研究 被引量:6

Study on Anti-Inflammatory Effect and Mechanism of 4'-O-β-D-glucosyl-5-O-methylvisamminol in Fangfeng(Saposhnikoviae Radix)

摘要 目的研究防风中5-O-甲基维斯阿米醇苷的抗炎作用及其机制。方法建立脂多糖(LPS)诱导RAW264.7小鼠单核巨噬细胞体外炎症模型,采用酶联免疫吸附测定法细胞中的白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、Toll样受体-4(TLR4)含量;采用葡聚糖硫酸钠(DSS)诱导小鼠结肠炎模型,观察5-O-甲基维斯阿米醇苷对结肠炎小鼠结肠损伤的影响,采用酶联免疫吸附测定法测定小鼠结肠组织上清液中丙二醛(MDA)和超氧化物歧化酶(SOD)水平,采用蛋白免疫印迹法(Western blot)检测结肠组织中肿瘤坏死因子(TNF-α)、IL-1β蛋白表达。结果与脂多糖组比,5-O-甲基维斯阿米醇苷各剂量组能降低RAW264.7炎症细胞中IL-1β、IL-6、TLR4含量(P<0.01),并呈现浓度依赖性;与空白组比,结肠炎模型组小鼠体质量及结肠长度降低,结肠组织有明显的病理改变。与模型组比,5-O-甲基维斯阿米醇苷各剂量组能缓解结肠炎小鼠肠炎症状和减轻肠黏膜病理损伤,降低结肠中MDA含量,增加SOD活性(P<0.01),并能下调结肠组织中的TNF-α、IL-1β蛋白表达水平(P<0.01)。结论防风中5-O-甲基维斯阿米醇苷可抑制LPS诱导小鼠RAW264.7细胞炎症,对DSS诱导小鼠结肠炎有一定的治疗作用。 Objective To study the anti-inflammation effect and mechanism of 4'-O-β-D-glucosyl-5-O-methylvisamminol in Fangfeng(Saposhnikoviae Radix).Methods The inflammatory cell model was established using LPS-induced RAW 264.7 cells.The contents of interleukin-1β(IL-1β),interleukin-6(IL-6)and Toll like receptor 4(TLR4)in the supernatant of RAW 264.7 cells inflammation model were determined by ELISA.By means of dextran sodium sulfate(DSS)-induced ulcerative colitis model,the colon mucosal lesions were observed.The levels of malondialdehyde(MDA)and superoxide dismutase(SOD)in the supernatant of mouse colon tissue were measured by ELISA and protein expressions of tumor necrosis factor-α(TNF-α)and IL-1βwere detected by Western blot.Results Compared with the LPS group,4'-O-β-D-glucosyl-5-O-methylvisamminol could significantly reduce the release of IL-1,IL-6 and TLR4 in the supernatantin with a dose-dependent manner(P<0.01).Compared with those of the blank group,the body weight and colon length of mice in colitis model group decreased and there were significant pathological changes in colon tissue.Compared with the model group,5-o-methylvisamicin can alleviate the symptoms of colitis and reduce the pathological damage of intestinal mucosa in mice with colitis,reduce the content of MDA in colon,increase the activity of SOD(P<0.01)and inhibit the protein expression levels of TNF-αand IL-1βin colon tissue(P<0.01).Conclusion 4'-O-β-D-glucosyl-5-O-methylvisamminol in Fangfeng(Saposhnikoviae Radix)can inhibit the inflammation of RAW264.7 cells induced by LPS and it has a certain therapeutic effect on DSS-induced colitis in mice.
