摘要
目的:明确防风主要化学成分组成,预测其质量标志物。方法:采用UPLC-Q-TOF-MS/MS技术和GC-MS技术分析和表征不同产地防风样品中的共有化学成分,进一步通过网络药理学筛选出对疾病贡献值高的化合物作为潜在质量标志物,最后结合多元统计方法分析出显著差异成分作为防风质量标志物。结果:分析和表征出防风中36种化合物结构,进一步筛选出10个潜在质量标志物。最后筛选出含量差异大的5-O-甲基维斯阿米醇苷、3′-O-乙酰基亥茅酚、升麻素苷作为质量标志物。此外防风挥发油中人参炔醇含量占比高,药理活性显著,且在各产地间存在变量差异,也应作为防风质量标志物。结论:该研究通过液质、气质联用技术快速全面分析得到防风非挥发性及挥发性共有成分,并结合网络药理学和多元统计分析方法预测质量标志物,此研究思路可为预测中药材质量标志物提供借鉴。
Objective: To determine the main chemical compositions of Saposhnikoviae Radix and to predict its quality markers.Methods: UPLC-Q-TOF-MS/MS and GC-MS techniques were used to analyze and identify the common chemical components in Saposhnikoviae Radix from different areas.Further, the compounds with high disease contribution value were selected as potential quality markers through network pharmacology.Finally, the significant difference components were analyzed as quality markers in the samples by multivariate statistical method.Results: The structures of 36 compounds in Saposhnikoviae Radix were analyzed and characterized, ten potential quality markers were further screened.Finally, 5-O-methylvisammioside, 3′-O-acetylhamaudol and prim-O-glucosylcimifugin with high content difference were selected as quality markers.In addition, the content of panaxynol in the volatile oil of Saposhnikoviae Radix was high, the pharmacological activity was significant, and it had variable differences among different producing areas, so it should be used as a quality marker of Saposhnikoviae Radix.Conclusion: In this study, the non-volatile components and common volatile components of Saposhnikoviae Radix are quickly and comprehensively analyzed by LC-MS and GC-MS,and the quality markers of Saposhnikoviae Radix are predicted by combining network pharmacology and multivariate statistical analysis methods.This research idea can provide reference for predicting the quality markers of traditional Chinese medicine.
