摘要
目的:探讨TMPRSS2在结直肠癌的表达及临床意义,并预测其调控结直肠癌的可能机制。方法:基于TIMER、UALCAN、GEO、cBioPortal、DNMIVD、Kaplan⁃Meier Plotter、Oncolnc、GENT2、LinkedOmics、Cytoscape和GDSC多个数据库或软件,分析TMPRSS2在结直肠癌的表达、突变情况和甲基化状态,分析其与结直肠癌预后、临床特征、免疫浸润及药物敏感性的关系。结果:TMPRSS2在结直肠癌中较正常结直肠组织的表达水平及甲基化水平显著降低,基因突变率占比2%。TMPRSS2高表达与直肠癌预后不良相关,但对结肠癌预后无影响。结肠癌中TMPRSS2表达水平与免疫细胞浸润水平呈正相关。TMPRSS2表达与SLC37A1、GPA33、TJP3、EPCAM等基因紧密相关,它们主要富集在肠吸收及细胞粘附等生物过程。此外,TMPRSS2的高表达与TW37、FK866、Camptothecin等多种药物耐药相关。结论:TMPRSS2在结直肠癌的发生发展中发挥重要作用,有望成为直肠癌患者预后的一种新型标记物。
Objective:To investigate the TMPRSS2 expression in colorectal cancer and its clinical relevance,and to predict the probable mechanism underlying its regulatory effect in colorectal cancer.Methods:By using several databases or software packages including TIMER,UALCAN,GEO,cBioPortal,DNMIVD,Kaplan⁃Meier Plotter,Oncolnc,GENT2,LinkedOmics,Cytoscape and GDSC,we analyzed the expression,mutation and methylation of TMPRSS2 in colorectal cancer,as well as its relationship with prognosis,clinical features,immune infiltration and drug sensitivity of colorectal cancer.Results:The expression and methylation of TMPRSS2 in colorectal cancer were significantly lower than as found for normal colorectal tissues,with a gene mutation rate of 2%.High TMPRSS2 expression was associated with poor prognosis in rectal but not in colon cancers.The expression level of TMPRSS2 in colon cancer was positively correlated with the level of immune cell infiltration.The expression of TMPRSS2 was closely related to a few genes including SLC37A1,GPA33,TJP3,and EPCAM which were mainly enriched in some biological processes such as intestinal absorption and cell adhesion.In addition,high TMPRSS2 expression was associated with resistance to a variety of drugs such as TW37,FK866,and Camptothecin.Conclusion:TMPRSS2 plays an important role in the occurrence and development of colorectal cancer,and is promising as a novel marker for prognosis in patients with colorectal cancer.
