摘要
目的 采用响应面法优化超声辅助提取番木瓜叶多酚的工艺,并对其抗氧化活性进行研究。方法 根据单因素试验的结果,选取影响提取番木瓜叶多酚含量较大的因素,采用响应面法优化其提取工艺;利用DPPH法和铁氰化钾法研究番木瓜叶多酚的抗氧化能力。结果 番木瓜叶中多酚的最佳提取工艺参数为乙醇浓度53%,液料比16 ml/g,提取时间124 min,超声功率90 W,番木瓜叶提取物中多酚含量为12.29 mg/g,该值与预测值(12.33 mg/g)相近。此外,体外抗氧化活性研究结果表明,番木瓜叶多酚对DPPH自由基的清除能力达90.92%,番木瓜叶多酚浓度为1.0 mg/ml时,其铁离子还原能力与1.0 mg/ml维生素C溶液相近。结论 响应面法优化得到的超声辅助提取番木瓜叶多酚工艺较为可靠,得到的多酚提取物显示了良好的抗氧化活性,为番木瓜叶的深入开发利用提供了一定的研究依据。
Objective To optimize the ultrasonic assisted extraction of polyphenols from papaya leaves by response surface methodology,and study the antioxidant activity of polyphenols.Methods According to the results of single factor experiment,the factors affecting the extraction of polyphenols from papaya leaves were selected,and the extraction process was optimized by response surface methodology.The antioxidant capacity of polyphenols from papaya leaves was determined by DPPH method and potassium ferricyanide method.Results The optimum extraction parameters of polyphenols from papaya leaves were as follows:the ethanol concentration was 53 %,the ratio of liquor to material was 16:1 ml/g,the extraction time was 124 min,and the ultrasonic power was 90 W.The content of polyphenols in the extract from papaya leaves was 12.29 mg/g,which was close to the predicted value of 12.33 mg/g.In addition,the antioxidant activity in vitro showed that the scavenging ability of papaya leaf polyphenols to DPPH radical was 90.92 % and the reducing ability to iron ion was 2.773.Conclusion The optimized ultrasonic assisted extraction of polyphenols from papaya leaves by response surface methodology is reliable.The obtained polyphenol extract shows good antioxidant activity,which can provide a certain research basis for further development and utilization of papaya leaves.
