一测多评法同时测定艾叶配方颗粒中6个酚酸类成分 被引量:5

Simultaneous determination of 6 phenolic acids in Artemisiae Argyi Folium formula granules by QAMS

摘要 目的建立一测多评(QAMS)法测定艾叶配方颗粒中6个酚酸类成分的含量,并验证其准确性及可行性。方法采用UPLC法,色谱柱为Waters ACQUITY UPLC HSS T3(150 mm×2.1 mm,1.8μm);流动相为乙腈-0.1%甲酸溶液,梯度洗脱;流速为0.3 mL·min^(-1);检测波长为325 nm。以绿原酸为内参物,计算新绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C的相对校正因子(?)。同时采用外标法和QAMS法测定艾叶配方颗粒中6个酚酸类成分的含量,比较2种方法的测定结果是否存在差异。结果新绿原酸、隐绿原酸、异绿原酸B、异绿原酸A、异绿原酸C在各自质量浓度范围内与峰面积线性关系良好(r≥0.999);?分别为1.00、0.94、1.20、1.25、1.16。QAMS法与外标法测得13批样品中6个酚酸类成分的含量结果无显著差异,相对误差均小于1.2%。结论建立的QAMS法测定结果准确,可用于艾叶配方颗粒的定量分析及质量控制。 Objective To establish a quantitative method to analyze simultaneously 6 phenolic acids in Artemisiae Argyi Folium formula granules by quantitative analysis of multi-components by single marker(QAMS),and to verify its accuracy and feasibility.Methods The chromatographic column was Waters ACQUITY UPLC HSS T3 (150 mm×2.1 mm,1.8μm),the mobile phase was acetonitrile-0.1% formic acid solution with gradient elution,the flow rate was 0.3 mL·min^(-1),and the detection wavelength was 325 nm.Chlorogenic acid was used as the internal reference material,and the relative correction factors of neochlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C were calculated.The content of 6 phenolic acids was determined by the external standard method and QAMS respectively.The QAMS was validated through comparison of the results obtained by both methods.Results Neochlorogenic acid,cryptochlorogenic acid,isochlorogenic acid B,isochlorogenic acid A and isochlorogenic acid C all had good linearity in their respective ranges (r≥0.999),and their relative correction factors were 1.00,0.94,1.20,1.25 and 1.16,respectively.The content of the 6 phenolic acids in 13 batches of samples by both method had no significant difference,with relative error was less than 1.2%.Conclusion The method is feasible and accurate,and can be used in the quantitative analysis and quality control.
