摘要
目的:建立一种超高效液相色谱串联质谱同时测定鸡肉、鸡蛋中尼卡巴嗪、金刚烷胺、金刚乙胺残留量的检测方法。方法:样品用0.1%乙酸乙腈提取,加入无水硫酸钠和正己烷萃取,氮吹浓缩后经PSA净化,通过C18色谱柱分离,采用0.1%甲酸水-乙腈梯度洗脱,多反应监测模式,金刚烷胺、金刚乙胺采用电喷雾正离子(ESI+)扫描模式,尼卡巴嗪采用电喷雾负离子(ESI-)扫描模式,内标法定量。结果:3种物质在1~100 ng·mL^(-1)浓度下呈良好线性关系,相关系数均大于0.998,方法定量限均为0.5μg·kg^(-1)。样品分别在0.5μg·kg^(-1)、1.0μg·kg^(-1)、5.0μg·kg^(-1)加标量下,回收率为80.4%~112.3%。结论:本方法可简便快捷同时测定尼卡巴嗪、金刚烷胺、金刚乙胺3种物质,且灵敏度高、准确度好,可节省成本。
Objective: To establish an ultra performance liquid chromatography series mass spectrometry for the simultaneous determination of nicabazine, amantadine and rimantadine residues in chicken and eggs. Method: The sample was extracted from 0.1% acetonitrile acetate, anhydrous sodium sulfate and n-hexane extraction, nitrogen blowing and then purified by PSA, separated by C18column, 0.1% water-acetonitrile gradient elution, multi-reaction monitoring mode(MRM), amantadine and diamond ethylamine were normalized by electric spray. In the sub(ESI+) scanning mode,nicabayazine adopts electrospray anion(ESI-) scanning mode, which is quantitative by internal standard method. Result:Experiments show that the three substances have a good linear relationship at a concentration of 1~ 100 ng·mL^(-1), with correlation coefficients greater than 0.997, and the quantitative limit of the method is 0.5 μg·kg^(-1). The recovery rate of the sample is 80.4%~112.3% in the range of 0.5 μg·kg^(-1), 1.0 μg·kg^(-1)and 5.0 μg·kg^(-1)respectively. Conclusion: This method can easily and quickly determine the three substances of nicabazine, amantadine and amantadine at the same time. With high sensitivity and accuracy, it can save costs.
