槲皮素介导IRF8/IFN-γ改善Hp感染诱导慢性萎缩性胃炎的机制研究 被引量:11

Mechanisms of quercetin-mediated IRF8/IFN-γin improving chronic atrophic gastritis induced by Hp infection

摘要 目的探究槲皮素介导干扰素调节因子8/γ干扰素(IRF8/IFN-γ)改善幽门螺杆菌(Hp)感染诱导慢性萎缩性胃炎(CAG)的机制。方法45只SD大鼠随机分为空白组10只和CAG组35只,采用水杨酸钠+乙醇+Hp悉尼菌株1菌液法灌胃建立Hp性CAG大鼠模型。将建模成功的CAG组大鼠余下30只大鼠随机分为模型组、槲皮素组(25 mg/kg)和胃复春组(0.43 g/kg),每组10只。相应药物干预30 d后处死大鼠,观察各组大鼠胃黏膜病理学变化,测定大鼠胃分泌功能(胃液pH、分泌量及胃蛋白酶活性),采用酶联免疫吸附法检测血清胃泌素17(G-17)、白细胞介素17(IL-17)和趋化因子1(CXCL1),实时荧光定量聚合酶链反应(PCR)和蛋白印迹法检测胃组织中IRF8、IFN-γ和干扰素诱导蛋白3(IFIT3)表达。结果模型组、槲皮素组和胃复春组胃黏膜组织炎症评分、胃液pH、血清IL-17、CXCL1、胃组织IRF8、IFN-γ和IFIT3 mRNA及蛋白表达均高于空白组,胃液总量、胃蛋白酶活性和G-17均低于空白组(P<0.05);槲皮素组和胃复春组胃黏膜组织炎症评分、胃液pH、血清IL-17、CXCL1、胃组织IRF8、IFN-γ和IFIT3 mRNA及蛋白表达均低于模型组,胃液总量、胃蛋白酶活性和G-17均高于模型组(P<0.05)。结论槲皮素通过抑制IRF8/IFN-γ轴,降低胃部炎症反应,增强其胃分泌功能,从而改善Hp感染诱导的CAG。 OBJECTIVE To explore the mechanisms of quercetin-mediated interferon regulatory factor 8/interferon-γ(IRF8/IFN-γ)in improving chronic atrophic gastritis(CAG)induced by Helicobacter pylori(Hp)infection.METHODS A total of 45 SD rats were randomly divided into 10 rats of blank group and 35 rats of CAG group.The models of rats with Hp-induced CAG were established by intragastric administration of sodium salicylate+ethanol+Hp SS1 bacterial solution.The remaining 30 rats in the successful modeled CAG group were randomly divided into model group,the quercetin group(25 mg/kg)and Weifuchun group(0.43 g/kg),with 10 rats in each group.The rats were sacrificed after 30 days of intervention with corresponding drugs.The pathological changes of gastric mucosa in each group were observed.The gastric secretion function of rats(gastric pH,secretion volume,pepsin activity)was detected.The serum gastrin 17(G-17),interleukin 17(IL-17)and chemokine 1(CXCL1)were detected by enzyme-linked immunosorbent assay(ELISA).The expressions of IRF8,IFN-γand interferon-induced protein with tetratricopeptide repeat-3(IFIT3)were detected by real-time quantitative polymerase chain reaction(PCR)and Western blot.RESULTS The score of gastric mucosal tissue inflammation,pH in gastric juice,levels of serum IL-17 and CXCL1,mRNA and proteins of IRF8,IFN-γand IFIT3 in gastric tissues in model group,quercetin group and Weifuchun group were all higher than those in blank group,while the total volume of gastric juice,pepsin activity and G-17 level were lower than those in blank group(P<0.05).The score of gastric mucosal tissue inflammation,pH in gastric juice,levels of serum IL-17 and CXCL1,mRNA and proteins of IRF8,IFN-γand IFIT3 in gastric tissues in quercetin group and Weifuchun group were lower than those in model group,while the total volume of gastric juice,pepsin activity and G-17 level were higher than those in model group(P<0.05).CONCLUSION Quercetin reduced gastric inflammation response and enhanced gastric secretion function by inhibiting IRF8/IFN-γ,thereby improved Hp infection-induced CAG.
