基于p38MAPK/NF-κB信号通路探讨苏木提取物对ApoE^(-/-)小鼠动脉粥样硬化的机制研究 被引量:11

摘要 目的 观察苏木提取物对动脉粥样硬化模型小鼠主动脉中p38MAPK/NF-κB信号通路相关蛋白表达的影响,进而探讨苏木提取物抗AS的作用机制。方法 所有小鼠适应性饲养2周后,将40只ApoE^(-/-)小鼠随机分为,模型组、苏木提取物低、中、高剂量组,每组10只;10只相同遗传背景的C57BL/6J小鼠设为空白组,其中空白组小鼠给予普通饲料喂养,其余四组小鼠给予高脂饲料喂养8周进行模型复制,第9周开始灌胃,连续灌胃4周,12周后分离小鼠主动脉根部,制备石蜡切片,HE染色观察小鼠主动脉窦病理形态学变化;ELISA法检测小鼠血清中IL-1β、TNF-α、ICAM-1、VCAM-1水平;生化法检测小鼠血清中TC、TG、LDLC、HDL-C水平;Western blot法检测小鼠主动脉中p38MAPK、NF-κB蛋白的表达。结果 苏木提取物低、中、高剂量组够改善AS模型小鼠主动脉窦病理形态学改变;降低小鼠血清中IL-1β、TNF-α、ICAM-1、VCAM-1及TC、TG、LDL-C水平;并可下调小鼠主动脉中p38MAPK、NF-κB蛋白的表达。结论 苏木提取物能够通过抗炎、降血脂等途径发挥抗AS的作用,部分机制可能与抑制p38MAPK/NF-κB信号通路活化有关。 Objective To observe the effect of Sappanwood ethyl acetate extract on the expression of p38MAPK/NF-κB signaling pathway related proteins in the aorta of atherosclerosis model mice, and then to explore the anti-AS mechanism of Sappanwood ethyl acetate extract.Methods After all mice were bred adaptively for 2 weeks, 40 ApoE^(-/-)mice were randomly divided into model group, Sappanwood ethyl acetate extract low, medium, and high dose groups, with 10 mice in each group;10 C57BL/6J mice with the same genetic background were set as the blank group. The mice in the blank group were fed with ordinary feed, and the other four groups were fed with high-fat feed for 8 weeks to replicate the model. The experiment started by gavage from the 9thweek, and continued gavage for 4 weeks. After 12 weeks, the mouse aortic roots were separated, paraffin sections were prepared, and HE staining was used to observe the pathological changes of the mouse aortic sinuses;ELISA method was used to detect the levels of IL-1β, TNF-α, ICAM-1 and VCAM-1 in the serum of mice;Biochemical method was used to detect the levels of TC, TG, LDL-C and HDL-C in the serum of mice;Western blot method was used to detect the expression of p38MAPK and NF-κB proteins in the mice aorta.Results The low, medium and high doses of Sappanwood ethyl acetate extract could improve the pathological changes of the aortic sinus in AS model mice;reduce the levels of IL-1β, TNF-α, ICAM-1, VCAM-1, TC, TG, and LDL-C in the serum of mice;and down-regulate the expression of p38MAPK and NF-κB proteins in the mice aorta.Conclusion Sappanwood ethyl acetate extract exerts anti-AS effects through anti-inflammatory and reducing blood lipid pathways. Part of the mechanism may be related to inhibiting the activation of p38MAPK/NF-κB signaling pathway.
