摘要
目的探讨中药益木脑液对肝硬化肝性脑病大鼠脑蛋白的影响及机制。方法SD雄性大鼠72只,随机分为空白对照组10只,模型组62只,模型组采用50%四氯化碳联合硫代乙酰胺腹腔注射制备肝性脑病大鼠模型,造模成功后的45只SD大鼠随机分为模型组、乳果糖组和益木脑液低、中、高剂量组,每组9只。乳果糖组按5.63 g/kg给予乳果糖灌肠,益木脑液低、中、高剂量组分别以0.56、1.12、2.24 g/kg的剂量给予益木脑液灌肠,模型组及对照组给予等量蒸馏水灌肠,每日1次,连续给药14 d,测定各组大鼠血氨、血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)水平;ELISA法检测大鼠血浆中的5-羟色胺(5-HT)、去甲肾上腺素(NE);采用免疫组织化学法检测大鼠脑组织中GFAP、GABA蛋白的表达。结果与对照组比较,模型组大鼠血清ALT、AST与血氨水平明显升高,血浆中5-HT与NE水平显著升高,脑组织中GABA蛋白、GFAP蛋白表达明显上升;与模型组比较,益木脑液低、中、高剂量组血清ALT、AST、血氨水平均降低(P<0.05),益木脑液中、高剂量组血浆中5-HT水平显著降低(P<0.01);益木脑液高剂量组血浆中NE水平显著降低(P<0.01);益木脑液低、中、高剂量组脑组织中GABA蛋白、GFAP蛋白表达显著降低。结论中药益木脑液对肝硬化肝性脑病具有治疗作用,其机制可能与降低血氨、调节神经递质水平、相关GFAP、GABA神经递质受体的表达有关。
Objective:To investigate the effect and mechanism of Yimunaoye on the cerebral nerve of rats with hepatic encephalopathy caused by liver cirrhosis.Methods:A total of 72 male SD rats were randomly divided into blank control group(10 rats)and model group(62 rats).The model group used 50%carbon tetrachloride combined with intraperitoneal injection of thioacetamide to prepare a rat model of hepatic encephalopathy,and 45 SD rats after successful modeling were randomly divided into model groups,lactulose groups and Yimunaoye low,medium and high dose groups,9 rats in each group.The lactulose group was given lactulose enema at 5.63 g/kg,Yimunaoye low,medium and high dose groups were given Yimunaoye enema at doses of 0.56,1.12 and 2.24 g/kg respectively,and the model group and blank control group were given an equal volume of distilled water enema,once a day,administered continuously for 14 days.The blood ammonia,serum aspartate aminotransferase(AST)and alanine aminotransferase(ALT)levels of the rats in each group were measured;ELISA method was used to detect the rat 5-hydroxytryptamine(5-HT)and norepinephrine(NE)in plasma;GFAP and GABA protein expressions in rat brain tissue were detected by immunohistochemistry.Results:Compared with the control group,the serum ALT,AST and blood ammonia levels of the model group were significantly increased,the plasma 5-HT and NE levels were significantly increased,the GABA protein expression in the brain tissue was significantly increased,and the GFAP protein expression was significantly increased;compared with the model groups,the serum ALT,AST and blood ammonia levels in the Yimunaoye low,medium and high dose groups were all decreased(P<0.05),and the level of 5-HT in plasma in the Yimunaoye medium and high dose groups was significantly decreased(P<0.01);the level of NE in the plasma of the Yimunaoye high dose group was significantly decreased(P<0.01);the expression of GABA protein in the brain tissue of the Yimunaoye low,medium and high dose groups was significantly de-creased,and the expression of GFAP protein was significantly decreased.Conclusion:Yimunaoye has a therapeu-tic effect on hepatic encephalopathy of liver cirrhosis,and its mechanism may be related to lowering blood ammo-nia,regulating the level of the neurotransmitter,and the expression of related GFAP and GABA neurotransmitter re-ceptors.
