摘要
为探究不同品种彩色马铃薯主要花色苷提取物种类及其抗氧化活性,通过高效液相色谱-二级管阵列-串联质谱法(HPLC-DAD-MS/MS)对马铃薯花色苷提取物的主要结构进行鉴定分析,采用体外抗氧化方法(1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl,DPPH))自由基法、铁离子还原/抗氧化能力法(Ferric ion reducing antioxidant power,FRAP)、2,2’-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(2,2’-Azinobis-(3-Ethylbenzthiazoline-6-sulphonate acid),ABTS)自由基法对马铃薯花色苷提取物的抗氧化活性进行测定,并采用抗氧化活性综合(Antioxidant potency composite,APC)指数法进行比较分析。HPLC-DAD-MS/MS检测结果表明:R618和R15-4主要花色苷为天竺葵素-3-香豆酰-芸香糖苷-5-葡萄糖苷;P15-6主要花色苷为锦葵素-3-O-对香豆酰芸香糖苷-5-O-葡萄糖苷;P601和P4-3主要花色苷为矮牵牛素-3-O-对香豆酰芸香糖苷-5-O-葡萄糖苷。APC指数结果表明:花色苷成分的抗氧化活性为P4-3>P601>P15-6>R618>R15-4。证明紫薯的抗氧化活性优于红薯。
The objectives of this study were to analyze the kinds of anthocyanins extract from different varieties of potato and their antioxidant activities.The main architecture of anthocyanins extract from different varieties of potato was identification analysis by HPLC-DAD-MS/MS.The antioxidant activities were determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH),2,2’-azinobis (3-ethylbenzthiazoline-6)-sulphonic acid (ABTS) and ferric ion reducing/antioxidant power (FRAP) methods,and comparatively evaluated using antioxidant potency composite (APC) index.The results of HPLC-DAD-MS/MS detection showed that the main anthocyanins of R15-4 and R618 was geranium-3-coumaroyl-trucoside-5-glucoside;P15-6 was kaempferol-3-O-p-coumarin-5-O-glucoside;P601 and P4-3 were petunin-3-O-p-coumarin-5-O-glucoside.The results of APC indexes showed that the antioxidant activities of the anthocyanins components were P4-3>P601>P15-6>R618>R15-4.The antioxidant activity of purple potato was better than that of red potato.
