摘要
以TY-4番木瓜组培芽为试验材料,在以半强度Murashige和Skoog培养基为基本培养基,蛭石为支撑物的基础上,研究不同蔗糖浓度(0、5、20、40 g/L)、吲哚-3-丁酸浓度(2、4、6、8 mg/L)、间苯三酚浓度(0、10.0、14.9、19.9mg/L)对番木瓜生长状态和生根率及根系质量的影响。结果表明:将番木瓜组培芽在含蔗糖浓度为0,吲哚-3-丁酸(IBA)浓度为2 mg/L、间苯三酚浓度为0,并补充1/2强度的MS大量元素、MS微量元素、MS有机质,肌醇0.1 g/L、硫胺素0.4 mg/L和核黄素0.38 mg/L,支撑物为蛭石的培养基中培养时,体外芽最高生根率为95.2%,与最低的生根处理相比,高出38.1%,且茎基部形成愈伤组织较少。通过正交试验结果分析可知,番木瓜最佳的生根组合为:蔗糖浓度为0、吲哚-3-丁酸(IBA)浓度为2 mg/L、间苯三酚浓度为10.0 mg/L。本研究可为番木瓜体外繁殖生根技术提供理论参考。
Tissue cultured shoots of papaya variety“TY-4”were cultured on rooting mediums with the half-strength Murashige and Skoog medium as basic medium and vermiculite as the support,supplemented with sucrose at different concentrations (0,5,20,40 g·L^(–1)),indole-3-butyric acid (IBA) at 2,4,6,8 mg·L^(–1),phloroglucinol at 0,10.0,14.9,19.9 mg·L^(–1)to observe the effects of the three factors,sucrose,IBA and phloroglucinol,on the growth,rooting rate and root quality.An orthogonal experiment was arranged to optimize the concentrations of the sucrose,IBA and phloroglucinol in the rooting medium.The results showed that when papaya tissue cultured shoots were cultured on the rooting medium including 1/2 strength MS with vermiculite as support,supplemented with macro-elements,trace elements,organic matter,0.1 g·L^(–1)inositol,0.4 mg·L^(–1)thiamine and 0.38 mg·L^(–1)riboflavin at the presence of 0 sucrose,2 mg·L^(–1)IBA,and 0 phloroglucinol,the rooting rate of the shoots in vitro was upto 95.2%,which was 38.1%higher than that of the treatment with the lowest rooting rate,under which less calli were formed at the base of the shoots.The orthogonal experiment showed that the optimal combination of the three factors in the papaya rooting medium is sucrose at the concentration of 0,IBA at 2 mg·L^(–1),and phloroglucinol at 10.0 mg·L^(–1).
