黔产大青木中潜在抗奶牛乳房炎活性组分的提取工艺及抗氧化活性研究 被引量:6

摘要 试验旨在优化黔产大青木中潜在抗奶牛乳房炎活性组分——类叶升麻苷及木樨草苷的提取工艺,并研究其抗氧化活性。在单因素试验的基础上,结合Box-Behnken响应面法以类叶升麻苷与木樨草苷得率的总评"归一值"为指标,优化提取工艺;测定其对DPPH自由基、ABTS自由基、羟自由基的清除能力。结果显示,最佳提取工艺条件为粒度目数100目、乙醇浓度62%、液料比22 mL/g、提取3次、提取时间1.5 h、提取温度83℃。此条件下,类叶升麻苷与木樨草苷得率分别为9.62、2.25 mg/g;两者对DPPH自由基、ABTS自由基、羟自由基具有一定的清除能力。研究表明,提取工艺优化后大青木中潜在抗奶牛乳房炎活性组分得率较高,具有较好的抗氧化活性,可为后续抗奶牛乳房炎的饲料添加剂研发提供参考。 The experiment optimized the extraction process of potential active ingredient against mastitis—acteoside and luteoloside in Clerodendrum cyrtophyllum Turcz. distributed in Guizhou Daqingmu, and investigated their antioxidant activity. On the basis of single factor test, combined with Box-Behnken response surface method, the extraction process was optimized by taking ’normalized value’ of the yield of acteoside and luteoloside as the index. The scavenging abilities of DPPH·, ABTS·and OH·were measured. The results showed that the optimum extraction conditions were particle size of 100 mesh, ethanol concentration of 62%, liquid-solid ratio of 22 mL/g, extraction for three times, extraction time of 1.5 h and extraction temperature of 83 ℃. Under the condition, the yields of acteoside and luteoloside were 9.62, 2.25 mg/g,respectively. The two active ingredient had a good scavenging ability for DPPH·, ABTS· and OH·. The experiment indicates that the potential active ingredient against mastitis components in Clerodendrum cyrtophyllum Turcz. has a high yield and shows good antioxidant activity. The experiment provides a reference for subsequent research and development of against mastitis feed additives.
