乌鲁木齐市羊源沙门菌的污染调查 被引量:2

Investigation of Salmonella contamination from sheep in Urumqi

摘要 为了解乌鲁木齐市周边绵羊养殖农户的沙门菌污染及其耐药性和致病力,采集乌鲁木齐市周边5家农户绵羊源的285份样品,按照国家标准GB 4789.4-2016进行沙门菌的分离培养,结合PCR方法鉴定,检测了19种毒力基因的编码情况,采用琼脂稀释法测定了13种抗菌药物的耐药情况。共分离鉴定出62株沙门菌,分离率为24.03%,其中粪便、肛拭子、饲料、体表拭子的检出率依次为13.82%(10/78),30.21%(29/96),13.33%(2/15),30.43%(21/69);19种毒力基因检出了16种,有沙门菌毒力岛上的sipB(100.00%)、invJ(98.39%)、mogA(87.10%)、spiA(98.39%)、sifA(87.10%)、ssaQ(100.00%)、sseL(66.13%)、mgtC(80.65%)、araB(98.39%)、sopB(93.55%),毒力质粒上的spvA(16.13%)、spvR(16.13%),菌毛上的lpfC(87.10%)、bcf(66.13%)以及肠毒素stn(96.77%);菌株的耐药率依次为:阿莫西林-克拉维酸钾和四环素(93.5%)、氨苄西林(91.9%)、头孢噻呋(80.6%)、多黏菌素E(79.0%)、头孢曲松钠(69.4%)、头孢西丁钠(66.1%)、卡那霉素(64.5%)、庆大霉素(59.7%)、恩诺沙星(40.3%)、氯霉素和阿奇霉素(1.6%);沙门菌的多重耐药最高达到10耐,依次从高到低是8耐(23.3%)、9耐(21.7%)、10耐(16.7%)、5耐和6耐(11.7%)、7耐(8.3%)、3耐(5.0%)、4耐(3.3%)。乌鲁木齐市周边散农户羊场的沙门菌污染率较高,具有较高的致病性,耐药性现象严重。 The aim of this study to understand the Salmonella contamination,drug resistance and virulence gene distribution of sheep farmers around Urumqi.A total of 285 samples from sheep of five farmers around Urumqi were collected.Salmonella was isolated and cultured according to the national standard GB 4789.4-2016.Nineteen virulence genes were detected by PCR method and the drug resistance of 13 antibiotics was determined by agar dilution method.A total of 62 Salmonella strains were isolated and identified from 285 samples and the isolation rate was 24.03%.The detection rates of feces,anal swab,feed and body surface swab were 13.82%(10/78),30.21%(29/96),13.33%(2/15)and 30.43%(21/69),respectively.Sixteen of the 19 virulence genes were identified,with sipB(100.00%),invJ(98.39%),mogA(87.10%),spiA(98.39%),sifA(87.10%),ssaQ(100.00%),sseL(66.13%),mgtC(80.65%),araB(98.39%),sopB(93.55%)on Salmonella virulence island,spvA(16.13%)and spvR(16.13%)on virulence plasmid,lpfC(87.10%),bcf(66.13%)on pili and enterotoxin stn(96.77%).The drug resistance rates of the isolates to 13 antibiotics were as follows,Amoxicillin-clavulanate potassium(93.5%)and tetracycline(93.5%),ampicillin(91.9%),cefuroxime(80.6%),polymyxin E(79.0%),ceftriaxone sodium(69.4%),cefoxitin sodium(66.1%),kanamycin(64.5%),gentamicin(59.7%),enrofloxacin(40.3%),chloramphenicol(1.6%)and azithromycin(1.6%).The highest multidrug resistance of Salmonella reached 10 resistances,in order from high to low were 8 resistances(23.3%),9 resistances(21.7%),10 resistances(16.7%),5 resistances and 6 resistances(11.7%),7 resistances(8.3%),3 resistances(5.0%)and 4 resistances(3.3%).The Salmonella contamination rate of sheep farms around Urumqi was higher.These strains have high pathogenicity and serious drug resistance.
