摘要
目的基于糖尿病并发症探讨地锦草总黄酮(Total flavoneofEuphorbiaofficinalis,TFEH)对T2DM大鼠糖脂代谢、炎性因子及氧化应激方面的干预作用。方法随机选取10只大鼠为空白组,其余动物采用高脂高糖饲料喂养4周+腹腔注射链脲佐菌素(STZ,75 mg/kg)构建T2DM模型。成模动物分为模型组、盐酸二甲双胍组、地锦草总黄酮高剂量组、地锦草总黄酮低剂量组,每组各10只。各组动物给予相应药物灌胃,1次/d,连续给药6周,2周测定一次空腹血糖值。酶联免疫Elisa法测定血清中糖化血清蛋白(GSP)、胰岛素(INS)、胰岛素自身抗体(IAA)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、超氧化物气化酶(SOD)、丙二醛(MDA),总抗氧化能力(TAOC)、肿瘤坏死因子(TNF-α)、白介素-6(IL-6)、C反应蛋白(CRP)、脂联素(ADPN)、瘦素(LEP)等指标。各组动物取全血检测全血黏度。结果与模型组比较,地锦草总黄酮可不同程度降低模型动物血清空腹动态血糖、IAA含量,提高INS含量,降低血清TC、TG、LDL-C含量,升高HDL-C含量,降低全血黏度(P<0.05);氧化应激方面可增加模型动物血清中SOD、T-AOC含量,降低MDA含量(P<0.05);炎症因子方面,可有效降低模型动物血清中TNF-α、IL-6、CRP含量,升高ADPN、LEP含量(P<0.05)。结论地锦草总黄酮可有效改善T2DM大鼠糖脂代谢紊乱、降低炎症反应及氧化应激反应的发生,控制T2DM并发症的发生及发展。
Objective Based on the complications of diabetes to explore the effect of total flavone of euphorbia offici⁃nalis(TFEH)on the metabolism of glucose and lipid,inflammatory factors and oxidative stress of T2DM rats.Methods Ten rats were randomly selected as the blank group.The other rats were fed with high fat and high sugar diet for 4 weeks and got intraperitoneal injection of streptozotocin(STZ,75mg/kg)to construct T2DM model.The constructed models were divided into the model group,metformin hydrochloride group,high-dose TFEH group and low-dose TFEH group.The rats in each group were given corresponding drugs once a day for 6 weeks,and fasting blood glucose was measured once every two weeks.Enzyme-linked immunoelisa method was used to test glycosylated serum protein(GSP),insulin(INS),insulin autoantibody(IAA),total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C),superoxide gasifying enzyme(SOD),malondialdehyde(MDA),the total antioxidant capacity(T-AOC),tumor ne⁃crosis factor(TNF-α),interleukin-6(IL-6),C-reactive protein(CRP),adiponectin(ADPN),leptin(LEP)and so on.The whole blood viscosity was measured.Results Compared with the model group,TFEH can reduce the serum fasting dynam⁃ic blood glucose and IAA content,increase INS content,reduce TC,TG and LDL-C content,increase HDL-C content,and reduce the whole blood viscosity.As for the oxidative stress,TFEH can increase the SOD and T-AOC content,reduce the content of MDA in the model group.As for the inflammatory factors,TFEH can effectively reduce TNF-α,IL-6 and CRP content and increase ADPN and LEP content in the model group.Conclusion TFEH can effectively improve the disorder of glucose and lipid metabolism,reduce the occurrence of inflammation and oxidative stress,control the occurrence and de⁃velopment of complications of T2DM,which can effectively prevent and treat diabetes.
