地锦草总黄酮提取工艺优选及其抑制α-葡萄糖苷酶活性研究 被引量:6

Optimization of Extraction Technology for Total Flavonoids from Euphorbia Humifusa Willd. and Study on Its Inhibitory Effects on α-glucosidase

摘要 目的:优化地锦草总黄酮的超声提取工艺,并对其抑制α-葡萄糖苷酶活性进行研究。方法:在单因素实验基础上,运用Plackett-Burman(PB)试验设计筛选影响地锦草总黄酮提取量的主要因素,选用Box-Behnken设计优化地锦草总黄酮的超声提取工艺,以乙醇体积分数、液料比和超声功率为自变量,总黄酮提取量为因变量,通过对自变量与因变量的完全二次响应曲面的回归拟合,应用效应面法对最佳工艺进行预测分析;采用α-葡萄糖苷酶抑制模型,对地锦草总黄酮抑制α-葡萄糖苷酶的活性进行研究。结果:最佳提取工艺为乙醇体积分数82.60%,液料比15.95 m L·g^-1,超声功率169.40 W,超声时间60 min,超声温度40℃,超声次数2次,在此条件下地锦草总黄酮提取量为52.01 mg·g^-1,与预测值的相对误差为2.69%;对α-葡萄糖苷酶的半数抑制浓度(IC50)为1073.00μg·m L^-1。结论:优选的提取工艺简便、重复性好、预测性强,且地锦草总黄酮有较强的α-葡萄糖苷酶抑制活性。 Objective:To optimize the ultrasonic extraction technology of total flavonoids extraction from Euphorbia humifusa Willd.,and to investigate its inhibitory effects on α-glucosidase in vitro. Methods:Based on single factor test,Plackett-Burman test was adopted to screen main influencing factors;Extraction technology of total flavonoids from Euphorbia humifusa Willd. was optimized by Box-Behnken response surface methodolog,with the concentration of ethanol,solid-liquid ratio and ultrasonic power as independent variables,dependent variable was the yield of total flavonoids,through complete quadratic response surface regression fitting of independent variables and the dependent variable,optimum technology was determined by intuitive analysis of response surface methodology,and predictive analysis was used;α-glucosidase inhibition model in vitro were adopted to measure inhibition effect of total favonoids on α-glucosidase. Results:Optimum extraction technology was as following:ethanol concentration of 82.60%,solid-liquid ratio of 1∶15.95,ultrasonic power of 169.40 W,ultrasonic temperature of 40.00 ℃,ultrasonic time of 60.00 min for two times,under these conditions,the yield of total flavonoids was 52.01 mg·g^-1,and the relative error with the predicted value was 2.69%. Inhibitory effects of total flavonoids to α-glucosidase with IC50 of 1073.00 μg·m L^-1. Conclusion:The ultrasonic extraction technology was verified as simple,accurate,reproducible with good predictability,and total flavonoids show significant α-glucosidase inhibitory activities.
