电针天枢对肠易激综合征模型大鼠结肠敏感性与心率频域的调控作用 被引量:10

Regulation effect of electroacupuncture at Tianshu on the colonic sensitivity and the heart rate frequency domain in IBS model rats

摘要 目的从自主神经平衡性的角度探索单独电针天枢穴对IBS模型大鼠结肠敏感性的调节机制。方法选取体质量200~250 g的雄性清洁级Wistar大鼠36只,随机分为正常对照组、模型组、电针天枢组。采用避水应急法造模。造模结束后运用腹壁撤回反射(AWR)半定量评分对模型进行评价。选用天枢,毫针直刺0.3 cm后接电针诊疗仪,参数为连续波、2 Hz、1 mA,连续20 min,每日1次,连续14 d。对照组和模型组每天与天枢组相同时间相同方法进行抓取、固定,但不给予电针干预。内脏敏感性采用腹壁撤回反射(AWR)测定,运用Powerlab数据采集分析系统,记录大鼠在不同扩张压力下对应的腹直肌放电活动。使用Powerlab数据采集分析系统记录标准Ⅱ导联心电图,分析其变异性。结果与模型组比较,电针天枢治疗后,IBS大鼠的内脏疼痛阈值明显升高(P<0.05),LF、LF/HF显著降低(P<0.01、P<0.001)。结论单纯电针天枢对IBS模型大鼠VHS与自主神经系统平衡性均有明显的调控作用,提示电针天枢对IBS模型大鼠自主神经系统平衡性的修复作用可能是电针天枢调节VHS的机制之一。 Objective To explore the regulation mechanism of the electroacupuncture at Tianshu(S25)on the colonic sensitivity of IBS model rats from the perspective of autonomic nervous balance.Methods A total of 36 clean male Wistar rats weighing 200-250 g were selected and randomly divided into a normal control group,a model group,and an electroacupuncture at Tianshu group.The model was made by the closed water test and was evaluated by the semi-quantitative score of abdominal withdrawal reflex(AWR).An EA apparatus was connected after needling 0.3 cm perpendicularly by filiform needle at Tianshu.The parameters were continuous wave with 2 Hz and 1 mA for 20 minutes.The treatment was given once every day and continued for 14 days.The control group and model group were grabbed and fixed with the same method at the same time every day as the Tianshu group,but they were not given the EA intervention.The visceral sensitivity was measured by the abdominal withdrawal reflex(AWR),and the corresponding rectus abdominis discharge activities of the rats under different dilation pressures were recorded by the Powerlab data acquisition and analysis system.The standard Ⅱ lead ECG was recorded and its variability was analyzed by the system,too.Results Compared with the model group,the visceral pain threshold of IBS rats was significantly increased after the electroacupuncture treatment at Tianshu(P<0.05),while the LF,LF/HF were significantly decreased(P<0.01,P<0.001).Conclusion The electroacupuncture at Tianshu can significantly regulate the balance of the VHS and the autonomic nervous system in IBS model rats,suggesting that the repair effect of the electroacupuncture at Tianshu on the balance of the autonomic nervous system in IBS model rats may be one of the mechanisms on regulating(visceral hypersensitivity,VHS)with the electroacupuncture at Tianshu.
