摘要 目的研究microRNA-21及其调控基因EGFR在非小细胞肺癌-肺腺癌和鳞癌中的表达及意义。方法以2020年10月至2020年12月于昆明医科大学第一附属医院就诊的30例经病理确诊的非小细胞肺癌患者的肺组织标本作为研究对象,包括肺腺癌24例和肺鳞癌6例,同时以癌旁肺组织作为对照。实时定量PCR检测microRNA-21的表达,免疫组织化学方法检测microRNA-21调控基因EGFR的蛋白表达和定位。结果癌旁肺组织中microRNA-21和EGFR表达水平较低,与癌旁组织比较,肺腺癌和肺鳞癌组织中microRNA-21和EGFR表达均明显上调,差异有统计学意义(P<0.01);与肺腺癌组织比较,肺鳞癌组织中的microRNA-21和EGFR表达明显升高,差异有统计学意义(P<0.05)。结论microRNA-21及其调控基因EGFR在非小细胞肺癌-肺腺癌和鳞癌中呈高表达,且表达趋势一致,可预测非小细胞肺癌的进展与预后。 Objective To explore the expression and significance of microRNA-21 and its regulatory gene EGFR in non-small cell lung cancer,lung adenocarcinoma and squamous cell carcinoma. Methods Lung tissue samples of 30 patients with pathologically confirmed non-small cell lung cancer from October 2020 to December2020 in The 1 st Affiliated Hospital of Kunming Medical University were taken as the research objects,including 24 cases of lung adenocarcinoma and 6 cases of lung squamous cell carcinoma,and the lung tissue adjacent to cancer was taken as the control. The expression of microRNA-21 was detected by real-time quantitative PCR,and the protein expression and localization of EGFR were detected by immunohistochemistry. Results The expression levels of microRNA-21 and EGFR in lung tissue adjacent to cancer were lower. Compared with lung tissue adjacent to cancer, the expression levels of microRNA-21 and EGFR in lung adenocarcinoma and lung squamous cell carcinoma were significantly increased(P < 0.01);compared with lung adenocarcinoma,the expression levels of microRNA-21 and EGFR in lung squamous cell carcinoma were significantly increased( P < 0.05).Conclusion MicroRNA-21 and its regulatory gene EGFR are highly expressed in NSCLC lung adenocarcinoma and squamous cell carcinoma,and the expression trend is consistent,which can be used to predict the progress and prognosis of NSCLC.
