摘要
为揭示泡桐抗逆机制,以8 a生白花泡桐的形成层和韧皮部为试验材料,利用高通量测序技术Illumina研究了二倍体和同源四倍体抗逆相关基因的差异表达变化。测序结果表明,二、四倍体泡桐的Clean reads与泡桐参考基因组序列的比对率分别为91.47%~91.79%和92.08%~92.50%,且测序质量较好。可变剪接预测分析发现,外显子可变剪切数量最多,且四倍体泡桐中的可变剪切异构体多于二倍体。抗逆性相关基因的筛选研究结果中共鉴定到3138个与抗逆性相关的差异基因(1929个上调,1209个下调),并对其进行GO功能和KEGG代谢通路分析。采用qRT-PCR技术对随机8个差异基因进行检测,结果显示趋势均与测序结果一致。为了进一步研究抗逆相关基因的功能,对泡桐和其他11个物种的多酚氧化酶(PPO)基因序列进行了进化树分析,结果表明,泡桐与其他物种的PPO基因差异较大,表明泡桐可能是通过PPO基因倍增和丢失的差异变化来响应逆境胁迫。
To reveal the stress resistance mechnaism of Paulownia,Illumina high-throughput sequencing technology Illumina was used to study the differential expression changes of genes related to stress resistance between diploid and autotetraploid in cambium and phloem tissues of 8-year-old Paulownia fortunei.The sequencing results showed that the alignment rates of clean reads to the Paulownia reference genome sequences about diploid and autotetraploid were 91.47%~91.79%and 92.8%~92.50%,respectively,and the sequencing quality was good.Bioinformation analysis of alternative splicing showed the number of variable splicing in exons was the largest,and there were more variable splicing isomers in autotetraploid Paulownia.A total of 3138 differentially epressed genes related to stress resistance(1929 up-regulated and 1209 down-regulated)were identified,and GO function and KEGG pathway analyses were performed.QRT-PCR was used to detect 8 differentially epressed genes,which were selected randomly,and the results showed that the trend was consistent with the sequencing results.In order to further study the functions of genes related to stress resistance,phylogenetic tree analysis was performed on the polyphenol oxidase(PPO)genes sequences of Paulownia and other 11 species.The analysis results showed that PPO genes were quite different between paulownia and other species,indicating that paulownia may respond to adversity stress through differential changes in PPO gene doubling and loss.
