摘要
目的建立并评价大鼠激素性股骨头坏死模型。方法选择清洁级实验大鼠50只,按随机数表法将大鼠均分为实验组和对照组各25只。实验组采用注射脂多糖及大剂量甲基强的松建立大鼠激素性股骨头坏死模型,对照组同时间给予等量生理盐水注射,两组均观察6周后处死。比较两组大鼠活动情况指标,包括开场试验中活动平均速度、活动总距离及3 min内自主活动次数、最大抓力。分析两组大鼠生化指标变化,包括血清睾酮(T)、促甲状腺激素(TSH)、促肾上腺皮质激素(ACTH)及血浆环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)水平。观察两组组织病理学情况,包括骨小梁宽度、股骨头单位面积内骨小梁面积占比、每视野下骨空泡陷窝数量及单位面积骨小梁内胶原含量。结果实验组大鼠开场试验中活动平均速度、活动总距离及3 min内自主活动次数、最大抓力均低于对照组(P<0.05)。实验组大鼠T、TSH、ACTH、cAMP水平均低于对照组(P<0.05),cGMP水平高于对照组(P<0.05)。实验组大鼠骨小梁宽度、股骨头单位面积内骨小梁面积占比、骨小梁内胶原含量均低于对照组(P<0.05),空泡陷窝数量高于对照组(P<0.05)。结论采用脂多糖及大剂量甲基强的松可成功诱导大鼠激素性股骨头坏死模型。
Objective To establish and evaluate the model of steroid induced osteonecrosis of femoral head in rats.Methods Fifty clean SD rats were randomly divided into experimental group and control group with 25 rats in each group.The experimental group was injected with lipopolysaccharide and high-dose methylprednisone to establish the rat model of steroid induced femoral head necrosis,while the control group was injected with the same amount of normal saline at the same time.The activity indexes of the two groups were compared,including the average speed,the total distance,the number of independent activities and the maximum grasping force in 3 minutes,The changes of biochemical indexes including serum testosterone(T),thyroid stimulating hormone(TSH),adrenocorticotropic hormone(ACTH)and plasma cyclic adenosine monophosphate(cAMP)and cyclic guanosine monophosphate’(CGMP)levels were analyzed,The histopathological changes of the two groups were observed,including the width of trabecular bone,the ratio of trabecular area per unit area of femoral head,the number of bone vacuoles and lacunae per visual field and the content of collagen in trabecular bone per unit area.Results The average speed,total distance,number of independent activities and maximum grip in the experimental group were significantly lower than that of the control group(P<0.05).The levels of T,TSH,ACTH and camp in the experimental group were significantly lower than those in the control group(P<0.05),and the cGMP level was significantly higher than that in the control group(P<0.05).The width of trabecular bone,the ratio of trabecular area per unit area of femoral head and the content of collagen in trabecular bone of experimental group were significantly lower than those of control group(P<0.05),and the number of vacuole lacunae was significantly higher than that of control group(P<0.05).Conclusion The model of steroid induced necrosis of femoral head in rats is successfully induced by lipopolysaccharide and high-dose methylprednisone.
