奥沙利铂抗肿瘤及其耐药机制的研究进展 被引量:9

摘要 奥沙利铂是已被证明对结直肠肿瘤切实有效的铂类药物,并已成为该肿瘤的一线化疗方案用药,不仅如此,奥沙利铂也应用于胰腺癌及胃癌的辅助治疗。然而,此类肿瘤患者常出现治疗失败,主要是由于这类患者存在先天性或获得性耐药。因此,了解奥沙利铂的耐药机制对于克服这一现象并提高精准治疗水平非常重要。全文复习目前对于奥沙利铂肿瘤耐药机制的最新研究进展,包括药物摄入、转出、DNA损伤修复、细胞死亡、自噬在调控奥沙利铂耐药中的关键作用。 Oxaliplatin is a platinum drug that has been proven to be effective against colorectal tumors,and has become the first-line chemotherapy regimen in treatment of this tumor.Not only that,oxaliplatin is also used in the adjuvant treatment of pancreatic cancer and gastric cancer.However,treatment failure often occurs in patients with such tumors,which is mainly due to congenital or acquired drug resistance.Therefore,understanding the resistance mechanism of oxaliplatin is very important to overcome this phenomenon and improve the level of precision treatment.This paper reviews the latest research progress in the mechanism of oxaliplatin resistance,including the key role of drug intake,transfer,DNA damage repair,cell death,and autophagy in regulating oxaliplatin resistance.
