放血疗法对奥沙利铂所致周围神经毒性的缓解作用 被引量:1

摘要 目的观察放血疗法对大鼠奥沙利铂所致周围神经毒性的缓解作用。方法30只大鼠随机分为正常组、模型组和放血组。模型组和放血组隔天腹腔注射奥沙利铂溶液2 mg/kg,共4次,正常组注射等体积5%葡萄糖溶液。造模后第2天起,放血组予双后足井穴放血治疗,隔天1次,共10次。大鼠于造模前(d0)、造模结束后(d7)、第5次治疗后(d16)和第10次治疗后(d26)30 min,进行机械性痛觉敏化测试;第10次治疗后(d26),测定大鼠后足背皮肤血流灌注情况。结果造模前(d0)各组大鼠15 g、4 g VonFrey刺激阳性反应率差异均无统计学意义(P>0.05)。造模结束后(d7),15 g、4 g VonFrey刺激模型组和放血组阳性反应率均高于正常组(P<0.05),且模型组和放血组之间差异无统计学意义(P>0.05)。第5次治疗后(d16)和第10次治疗后(d26)后,模型组、放血组大鼠4 g、15 g Von-Frey刺激阳性反应率均高于正常组(P<0.05);放血组阳性反应率显著低于模型组(P<0.05)。第10次治疗后(d26),正常组大鼠后足背皮肤血流灌注多于模型组和放血组,放血组多于模型组。结论放血疗法可以降低大鼠奥沙利铂所致痛觉过敏和超敏反应,提高皮肤血流灌注而缓解周围神经毒性。 Objective To observe the relieving effects of bloodletting therapy on oxaliplatin-induced peripheral neurotoxicity in rats.Methods Thirty rats were randomly divided into the normal group,model group and bloodletting group.The model group and bloodletting group were treated with intraperitoneal injection of oxaliplatin solution at 2 mg/kg every other day for 4 times,and the normal group was treated with 5%glucose solution at equal volume.From the second day after modeling,the rats in the bloodletting group were treated with bloodletting at Jing acupoint of both hind paws,once every other day for 10 times.The rats were performed the mechanical pain sensitization test before modeling(d0),after modeling(d7),after the 5 th treatment(d16),and after the 10 th treatment(d26).After the 10 th treatment(d26),the blood perfusion in skin of the hind paw was measured.Results Before modeling(d0),there was no statistically significant difference in the positive response rate of 15 g and 4 g VonFrey stimulation among these group(P>0.05).At modeling(d7),the positive reaction rate of 15 g and 4 g VonFrey stimulation in the model group and bloodletting group was higher than that of the normal group(P<0.05),and there was no statistically significant difference between the model group and bloodletting group(P>0.05).After the 5 th treatment(d16)and the 10 th treatment(d26),the positive response rate of 4 g and 15 g VonFrey stimulation in the model group and bloodletting group was higher than that in the normal group(P<0.05),and the positive response rate in the bloodletting group was significantly lower than that in the model group(P<0.05).After the 10 th treatment(d26),the blood perfusion of the normal group was more than that of the model group and the bloodletting group,and that of the bloodletting group was more than that of the model group.Conclusion Bloodletting therapy can reduce the hyperalgesia and hypersensitivity caused by oxaliplatin,improve the blood perfusion of skin and alleviate the peripheral neurotoxicity in rats.
