丹参花与根部多糖抗氧化活性及其组成研究 被引量:7

Analysis of Monosaccharide Composition and Antioxidant Activity of Salvia Miltiorrhiza and Salvia Miltiorrhiza Polysaccharides

摘要 目的提取纯化丹参花部多糖(SMFP)与根部多糖(SMRP),分析它们的抗氧化活性、结构与单糖组成,揭示丹参多糖的药用价值,为丹参植物资源的开发利用提供参考。方法通过脱脂、水提、醇沉获得多糖,以FRAP、Fenton及DPPH等方法测定多糖的抗氧化活性,用PMP-HPLC法分析多糖的单糖组成,通过使用红外光谱分析多糖结构。结果丹参花和丹参根粗多糖得率为7.48%、9.00%。SMFP总抗氧化能力最大为48.323 U/mL,羟自由基清除能力为64.1745 U/mL,DPPH清除率为90.57%,SMRP总抗氧化能力最大为39.335 U/mL,羟自由基清除能力为66.1310 U/mL,DPPH清除率为90.25%,阳性对照组1 mg/mL的Vc的总抗氧化能力为56.580 U/mL,羟自由基清除能力为76.6618 U/mL,DPPH清除率为97.12%;SMFP与SMRP均为β-吡喃多糖,SMFP的单糖组成摩尔比为甘露糖(Man)∶鼠李糖(Rha)∶葡萄糖醛酸(Glc UA)∶半乳糖醛酸(Gal UA)∶葡萄糖(Glc)∶半乳糖(Gal)∶阿拉伯糖(Ara)=1.7216∶1∶0.5524∶0.2275∶20.8683∶36.1826∶8.0239,SMRP单糖组成摩尔比为Man∶Rha∶Glc∶Gal=0.63∶1∶1.25∶8.32。结论丹参花与根部的多糖均有较强的抗氧化活性;SMFP与SMRP均为β-吡喃多糖,SMFP单糖组成更为丰富;根据单糖组成推测SMFP应具有抗肿瘤、降血糖等活性,较强的抗氧化活性也是丹参多糖保护神经细胞、抑制黑色素生成、保肝等活性基础。该研究可为丹参多糖药理深化研究及丹参植物资源的综合利用提供参考。 Objective Extraction and purification of Salvia miltiorrhiza polysaccharides(SMFP)and root polysaccharides(SMRP),analysis of their antioxidant activity,structure and monosaccharide composition,revealing the medicinal value of Salvia miltiorrhiza polysaccharides,and providing references for the development and utilization of Salvia miltiorrhiza plants.Methods In this study,polysaccharides were obtained by degreasing,water extraction,and alcohol precipitation.The antioxidant activities of polysaccharides were determined by FRAP,Fenton,and DPPH.The monosaccharide composition of the polysaccharides was analyzed by PMP-HPLC.Finally,the structure of the polysaccharides was analyzed by infrared spectroscopy.Results The yields of crude polysaccharides from Salvia miltiorrhiza flowers and Salvia miltiorrhiza roots were 7.48%and 9.00%.The total antioxidant capacity of SMFP was 48.323 U/mL,the hydroxyl radical scavenging ability was 64.1745 U/mL,and the DPPH scavenging rate was 90.57%.The SMRP total antioxidant capacity is 39.335 U/mL,and the hydroxyl radical scavenging ability is 66.1310 U/mL,DPPH clearance rate was 90.25%;total antioxidant capacity of positive control group(1 mg/mL Vc)was 56.580 U/mL,hydroxyl radical scavenging capacity was 76.6618 U/mL,DPPH clearance rate 97.12%;SMFP and SMRP are bothβ-pyran polysaccharides,and the molar ratio of the monosaccharide composition of SMFP was mannose(Man)∶rhamnose(Rha)∶glucuronic acid(Glc UA)∶galacturonic acid(Gal UA)∶glucose(Glc)∶galactose(Gal)∶arabinose(Ara)=1.7216∶1∶0.5524∶0.2275∶20.8683∶36.1826∶8.0239,the molar ratio of SMRP monosaccharide composition was Man∶Rha∶Glc∶Gal=0.63∶1∶1.25∶8.32.Conclusion Both SMFP and SMRP areβ-pyran polysaccharides,and both have strong antioxidant activity.It is speculated that the SMFP should have anti-tumor,hypoglycemic and other activities based on the composition of monosaccharides.The strong antioxidant activity also protects nerve cells and inhibits salvia polysaccharides.Basis for melanin production and liver protection.The research can provide reference for further research on pharmacological effects of Salvia miltiorrhiza polysaccharides and comprehensive utilization of Salvia miltiorrhiza plant resources.
