摘要
目的探讨SN在改善小鼠胶原性关节炎(CIA)关节症状、抑制体液免疫和细胞免疫方面与甲氨喋呤(MTX)的异同以及与MTX是否有叠加效应。方法以CIA为研究对象,观察不同剂量SN和SN+MTX治疗后,小鼠关节炎症的变化;ELISA法测定各组小鼠脾细胞分泌IFN-γ的能力和小鼠血清中抗Ⅱ胶原抗体的水平。结果无论大剂量还是小剂量应用SN治疗CIA,其改善炎症症状、抑制IFN-γ和抗Ⅱ型胶原抗体的能力均不及经典抗风湿药物———MTX。但SN与MTX联合治疗CIA,疗效优于其他治疗方案。结论与临床长期应用SN治疗RA的经验相似,将SN作为一种辅助治疗药物,与MTX等二线药物联合应用,可以达到更确切的治疗效果。
Objective To investigate the immunosuppression effects of sinomenine (SN) on the humoral and cellular immunity, as well as the curative effi-(cacy) of SN on collagen-induced arthritis (CIA) mouse. Methods The CIA mice were treated with SN or SN + MTX at different dosages and the changes after the treatment were observed. The IFN-γ-secreting ability of spleen lymphocytes and the level of anti-CⅡ antibodies in serum were assayed by ELISA. Results SN could suppress the cellular and humoral immunity responses to CⅡ and alter the sequential development of arthritis, but the curative efficacy of SN was inferior to MTX. SN in combination with MTX had a better curative efficacy on arthritis than SN or MTX did. Conclusion Similar to the clinical impact in RA patients, SN might be a useful drug for the assistant treatment. Treat with SN in combination with MTX would be more beneficial to RA patients.
