摘要
目的探讨白桦脂酸(betulinic acid,BA)对盐酸间氯苯哌嗪(m-chlorophenylpiperazine,mCPP)诱导的小鼠焦虑样行为的作用及其相关抗氧化应激机制。方法将24只健康雄性ICR小鼠随机分为对照组(羧甲基纤维素钠灌胃3 d+皮下注射生理盐水)、焦虑模型组(羧甲基纤维素钠灌胃3 d+皮下注射mCPP)、BA处理组(BA混悬液灌胃3d+皮下注射mCPP),每组8只。各组皮下注射后30 min进行开场自发活动、高架十字迷宫实验来评估小鼠焦虑水平,行为学实验结束后断头取脑,检测皮层及海马组织中丙二醛(malondialdehyde,MDA)含量、超氧化物歧化酶(superoxide dismutase,SOD)活性、谷胱甘肽过氧化物酶(glutathione peroxidase,GPX)活性。结果相比mCPP诱导的焦虑模型组,BA预处理组小鼠中央区路程百分比、中央区停留时间百分比均增加,开臂进入次数百分比和开臂内停留时间百分比均显著提高,差异有统计学意义(P<0.01)。相比对照组,mCPP诱导的焦虑模型组小鼠皮质和海马MDA含量升高,SOD、GPx活性降低,差异有统计学意义(P<0.05);相较于焦虑模型组,经BA预处理后小鼠皮质和海马MDA含量降低,SOD、GPx活性升高,差异均有统计学意义(P<0.05)。结论BA可以改善mCPP诱导的小鼠焦虑样行为,改善焦虑的机制可能与增加脑组织中SOD、GPx活性有关。
Objective To determine the effect of betulinic acid(BA)on m-chlorophenylpiperazine hydrochloride(mCPP)-induced anxiety-like behavior in mice and the underlying anti-oxidative stress mechanism.Methods ICR mice were divided into control,mCPP(1 mg/kg i.h.30 min before behavioral tests),and BA groups[BA 50 mg/(kg·d),i.g.for 3 days+mCPP(1 mg/kg i.h.30 min before behavioral tests)].Each group completed spontaneous activity and elevated plus maze(EPM)tests to evaluate mouse anxiety.After the behavioral tests,the head was decapitated and the brain tissue was separated.The content of malondialdehyde(MDA),the activity of superoxide dismutase(SOD),and the activity of glutathione peroxidase(GPX)was determined in the cerebral cortex and hippocampus.Results Compared with the anxiety group induced by mCPP,BA increased the central distance and central time ratios(P<0.05).The time ratio in the open arm(P<0.05)and the entries ratio in the open arm of the EPM test were also increased(P<0.01).Compared with the control group,the MDA levels in the cortex and hippocampus of mice were increased in the mCPP group(P<0.05),and the activities of SOD and GPx were decreased(P<0.05).BA reduced the levels of MDA in the cortices and hippocampi of mice(P<0.05),and increased SOD and GPx activities(P<0.05).Conclusion BA reduced the anxiolytic effect and the mechanism underlying improved anxiety may be related to increasing the activities of SOD and GPx in brain tissue.
