摘要
本实验通过设计正交实验,采用控制变量的方法,以一年生黑穗醋栗‘布劳德’嫩茎和种子为外植体进行试管苗增殖培养、生根培养及移栽驯化研究,建立黑穗醋栗‘布劳德’品种的组培快繁体。结果表明:以‘布劳德’茎段为外植体,用75%酒精,20%的次氯酸钠消毒5 min的消毒方式,成活率达83%;首次采用Anderson培养基培养黑穗醋栗试管苗,培养效果相对较好;生根培养过程中,利用暗培养培养试管苗1~2 d,生根率可达95%;移栽驯化时,试管苗在基质Ⅰ(蛭石∶珍珠岩=1∶1)中生长最好,移栽成活率达88.33%。本研究结果建立了黑穗醋栗‘布劳德’完整的快繁体系,为黑穗醋栗工厂化育苗与规模化种植提供技术参考。
In this study, by designing orthogonal experiment and controlling variables, the bud and seed of annual blackcurrant ’Brodtorp’ were used as explants, and the tissue culture of ’Brodtorp’ variety of blackcurrant was established through the selection of explants, the study of tube seedling proliferation culture, rooting culture and transplanting and acculturation. The results showed that The stem segment of ’Brodtorp’ was used as explants,and the survival rate reached 83% by using 75% alcohol and 20% sodium hypochlorite disinfection for 5 min.For the first time, Anderson medium was used to culture currant seedlings in vitro, and the culture effect was relatively good. In the rooting culture process, the rooting rate of the test-tube seedlings can reach 95% by using dark culture for 1~2 d. When transplanting domestication, tube seedlings in matrixⅠ(vermiculite: pearlite=1∶1) grow best, transplanting survival rate was 88.33%. The results of this study established a complete rapid-propagation system of blackcurrant ’browder’, providing a technical reference for the factory seedling and large-scale cultivation of blackcurrant.
