摘要
目的观察补肾活血方配伍二甲双胍对促排卵PCOS模型大鼠子宫内膜HOXA11、雌孕激素受体表达的影响,及对子宫内膜容受性的作用。方法将40只6周龄雌性SD大鼠随机分为空白对照组10只和PCOS建模组(来曲唑诱导法)30只。将建模成功的大鼠随机分为模型组10只、方剂组10只、克罗米芬组10只,分别予以1%CMC溶液、补肾活血方+二甲双胍+克罗米芬溶液、克罗米芬溶液,连续灌胃5 d,处死大鼠并取其子宫内膜。利用免疫组化法检测ER、PR的表达,利用免疫印迹法检测HOXA11的表达。结果(1)与正常组大鼠相比,模型组、方剂组、克罗米芬组子宫内膜上的ER和PR表达均增多(P<0.05);HOXA11表达却低于正常组(P<0.05);(2)与模型组相比,方剂组的ER、PR表达较弱(P<0.05),HOXA11的表达量明显增加(P<0.05);且方剂组HOXA11的表达量与正常组无明显差异(P>0.05)。结论补肾活血方配伍二甲双胍可以通过调节促排卵PCOS大鼠子宫内膜上雌孕激素受体含量,并增加HOXA11的表达来改善子宫内膜的容受性,增加妊娠成功几率。
Objective To observe the effect of reinforcing kidney and activating blood prescription combined with metformin on the expression of HOXA11,estrogen and progesterone receptor in endometrium of PCOS model rats.Methods 40 female SD rats with 6-week-old rats were randomly divided into control group(10 rats)and PCOS model group(30 rats).After successfully building the model,the PCOS rats were randomly divided into Model Group(N=10),Prescription Group(N=10)and Enclomiphene group(N=10)and given 1%CMC solution,BUSHEN huoxue recipe+Metformin Enclomiphene solution and Enclomiphene solution for 5 days,rats were killed and their endometrium was removed.The expression of ER and PR was detected by Immunohistochemistry and HOXA11 was detected by Western blotting.Results compared with normal group,the expression of Er and PR in endometrium of model group,Formula Group and enclomiphene group increased(P<0.05),while the expression of Hoxa11 was lower than that of normal group(P<0.05),the expression of ER and PR in the formula group was weak(P<0.05),and the expression of HOXA11 was increased(P<0.05),and the expression of HOXA11 in the formula group was not significantly different from that in the normal group(P>0.05).Conclusion The reinforcing kidney and activating blood prescription combined with metformin can improve the endometrial receptivity and pregnancy success by regulating the content of estrogen and progesterone receptors in endometrium of PCOS rats and increasing the expression of HOXA11.
