摘要
目的探讨养血安胎方通过转化生长因子-β(TGF-β)/Smad通路对肾虚血瘀型复发性流产的作用机制。方法建立肾虚血瘀型复发性流产小鼠模型,运用HE染色观察子宫内膜形态;蛋白质印迹法和实时荧光定量PCR检测空白对照组,模型对照组,养血安胎方高、中、低剂量组血管内皮生长因子(VEGF)、TGF-β/Smad通路的表达情况。结果与模型对照组比较,养血安胎方高剂量组的VEGF、TGF-β、Smad3蛋白表达均显著升高(P<0.05)。结论养血安胎方能通过调控TGF-β/Smad通路促进子宫内膜VEGF表达,其效果与剂量成正相关,提示养血安胎方治疗复发性流产可能与其增加血管通透性、改善微循环障碍、调节胎盘植入作用机制有关。
objective To investigate the mechanism of Yangxue Antai Recipe acting on recurrent spontaneous abortion with kidney deficiency and blood stasis via TGF-β/Smad pathway.Methods A mouse model of recurrent spontaneous abortion with kidney deficiency and blood stasis was established.HE staining was used to endometrial morphology, and western blot and real-time fluorescence quantitative PCR were utilized to detect the expression of vascular endothelial growth factor(VEGF) and TGF-β/Smad pathways in the normal group, model group and the high, medium and low dose groups.Results Compared with the model group, the expression levels of VEGF, TGF-β and Smad3 of the high-dose group of Yangxue Antai Recipe were significantly increased(P<0.05).Conclusion Yangxue Antai Recipe can promote the expression of VEGF in endometrium through TGF-β/Smad pathway, and its effect is positively correlated with the dose, suggesting that Yangxue Antai Recipe for recurrent spontaneous abortion may be associated with increased vascular permeability, improvement of microcirculation disorder and regulation of placenta implantation.
